Phylogenetic analysis of Eimeria tenella isolated from the litter of different chicken farms in Mymensingh, Bangladesh

Abstract: Background Eimeria tenella is the most pathogenic intracellular protozoan parasite of seven Eimeria species causing chicken coccidiosis around the world. This species is particularly responsible for caecal coccidiosis leading to serious morbidity–mortality and financial loss in poultry production. Methods The present study explored the genetic diversity of E. tenella. Litter slurry was collected from 18 broiler farms located in Mymensingh district, Bangladesh. Litter samples were processed for oocyst isolation… Show more

“…Notably, E. maxima and E. mitis were more closely related to each other than to E. praecox and E. acervulina , in contrast with findings based on the nuclear 18S and internal transcribed spacer (ITS) and mitochondrial cox 1 datasets [ 53 , 55 57 ]. Nevertheless, these analyses provided a consistent, robust phylogenetic resolution for the 33 isolates and their congeneric species in the genus Eimeria : a paraphyletic relationship was shared among the 10 chicken Eimeria species, in agreement with our genetic distance study herein and the results of morphological and molecular biology studies [ 6 , 56 , 58 60 ].

“…The isolated DNA samples were sent to the State Research Institute of Agricultural Microbiology (Pushkin, Russia) for analysis. 16S DNA libraries were composed using Ferier_F515 (5 -3 ) GTGCCAGCMGCCGCGGTAA and Ferier_R806 (5 -3 ) GGACTACVSGGGTATCTAAT primers [37], as described below.…”

“…The visual examination of the inner organs of dead chickens after autopsy elucidated an edema and inflammation of the small intestine, and particularly the caeca in all birds. DNA was isolated from the ileum digesta and analyzed by PCR with primers EtF (AATTTAGTCCATCGCAACCCT) and EtR (CGAGCGCTCTGCATACGACA) specific to ITS-1 of the ribosome cluster [37]. The PCR was positive in the samples from six of the seven chickens that died in the NC group and in the single dead chicken from the experimental group KS2 (Table 4).…”

“…In addition, this industry is responsible for around 31 percent of the overall meat production in the country. According to Alam et al (2022), the chicken industry on a worldwide scale is a notable sub-sector within the area of agriculture. This sub-sector is distinguished by rapid expansion as well as large contributions to nutrition and food security.…”
