Mushrooms species are well known to have a high potential for bioactive compounds for decades. In the era of modern technologies, it is possible to uncover and explore the hidden ethnopharmacological values, comprising complex bioactive compounds of substantial health benefits. Lepista sordida is an edible and medicinal mushroom, and its distributed from Europe to Southeast Asia. However, there was no prior study on the bioactive compounds of L. sordida from Indonesia. In this present study, ethanol extracts of L. sordida were obtained and their extract properties were determined by gas chromatography-mass spectrometry (GC-MS). A total of 13 different chemical compounds were found in L. sordida ethanol extracts through GC-MS analysis. 4-O-Methylmannose, n-Hexadecanoic acid, and cis-13-Octadecenoic acid were noted as the major compounds with area percentages of 10.09%, 8.19%, and 6.49%, respectively. The result of PASS Server analysis suggested that the ethanolic extract of L. sordida has the pharmaceutical effect to treat phobic disorder and skin damage, preventing blood clotting, anti-inflammatory, and treating several infections which cause by bacteria, protozoa, and viruses. Therefore, it is strongly suggested to conduct further bioassay to confirm the pharmaceutical properties of this mushroom.

As a widely used food preservative, methyl paraben was experimentally evidenced with serious hormonelike adverse effects. Herein, a high performance thin-layer chromatography platformed bioluminescent bioautography and image analysis for the selective quantifi cation and confi rmation of methyl paraben was proposed and validated in vinegar and coconut juice. First, the detectability of the bioautography to the analyte on different layer materials was estimated, revealing that normal silica gel was the best choice. After that, the liquid of sample extract and working solution were separated to overcome the background noises due to co-extracted matrices. The separation result was then coupled to the optimized bioautography, enabling instant and straightforward screening of the targeted compound. For accurate quantifi cation, bioluminescent inhibition pattern caused by the analyte was processed by image analysis, giving useful sensitivity (LOD > 16 mg/kg), precision (RSD < 10.1%) and accuracy (spike-recovery rate 76.9%-112.2%). Finally, the suspected result was confi rmed by determining its MS fi ngerprint, further strengthening the reliability of screening.

Isolation, purification, and structural elucidation of Stropharia rugosoannulata polysaccharides with hypolipidemic effect

Stropharia rugosoannulata is a widely grown edible mushroom with a high nutritional value. S. rugosoannulata polysaccharides is one of the most important bioactive components of S. rugosoannulata and has a wide range of activities. A S. rugosoannulata polysaccharides, named SRF-3, was derived from the S. rugosoannulata extraction by freeze-thaw combine with hot water extraction method, then prepareed with DEAE-cellulose column and Sephacryl S-200 HR gel column, and its hypolipidemic activity was determined. The structural characteristics of SRF-3 were analyzed by infrared spectral scanning (FT-IR), ultra-high performance liquid chromatography (UHPLC), acid hydrolysis, methylation analysis, nuclear magnetic resonance (NMR), and Gas Chromatography-Mass Spectrometer (GC-MS). SRF-3 is composed of mannose, galactose, methyl galactose and fructose with ratios of 16, 12, 58 and 12, respectively. In addition, the average relative molecular mass of SRF-3 is approximately 24 kDa. The main chain of SRF-3 is mainly composed of repeating α-D-1,6-Galp and α-D-1,6-Me-Galp units, with branches in the O-2 position of Gal. The structure is presumed to be a mannogalactan, with a small amount of t-β-D-Manp present as a side chain. Hypolipidemic activity assay showed that SRF-3 had good antioxidant and hypolipidemic effects in vitro, suggesting that SRF-3 have potential application in reducing liver fat accumulation.
