Although the influence of nanoparticles (NPs) on developmental processes is better understood, little is known about their impact on somatic embryogenesis (SE). This process involves changes in the direction of cell differentiation. Thus, studying the effect of NPs on SE is essential to reveal their impact on cell fate. This study aimed to examine the influence of gold nanoparticles (Au NPs) with different surface charges on the SE of 35S:BBM Arabidopsis thaliana, with particular emphasis on the spatiotemporal localization of pectic arabinogalactan proteins (AGPs) and extensin epitopes in cells changing the direction of their differentiation. The results show that under the influence of nanoparticles, the explant cells of 35S:BBM Arabidopsis thaliana seedling origin did not enter the path of SE. Bulges and the formation of organ-like structures were observed in these explants, in contrast to the control, where somatic embryos developed. Additionally, spatiotemporal changes in the chemical composition of the cell walls during the culture were observed. Under the influence of Au NPs, the following effects were observed: (1) explant cells did not enter the SE pathway, (2) the impacts of Au NPs with different surface charges on the explants were variable, and (3) the compositions of the analyzed pectic AGPs and extensin epitopes were diverse in the cells with different developmental programs: SE (control) and non-SE (treated with Au NPs).

Hairy roots are widely known as a biological system for the production of highly diverse biomolecules. β‐carotene – a precursor for vitamin A – is known to be an anti‐oxidant and anti‐gastric cancer and protection agent against cardiovascular disease, heart disease and stroke. β‐carotene has been chemically synthesised and consumed by humans. However, the chemical process often produces a by‐product that may be harmful to human health. Therefore, this study established a protocol to induce hairy roots (HRs) from a Vietnamese carrot variety and produce natural β‐carotene. The Rhizobium rhizogenes ATCC15834 harbouring Ri plasmid and a Vietnamese carrot variety were used as materials for genetic transformation and HR induction studies. The result showed that approximately 50 HR lines were obtained. Culture medium supplemented with 30 mg/L of sucrose that gave the highest biomass of HR was shown in carrot HR line 30, which had a doubling time of 6.5 days. The highest content of β‐carotene extraction, at 128 mg/100g hairy roots, was achieved with a ratio volume (v/v) of 2‐propanol and plant samples of 20:1, followed by two hours’ incubation with 2‐propanol at 60 °C. Our study reveals a highly efficient protocol for Vietnamese carrot hairy root establishment and multiplication. A very efficient protocol for β‐carotene extraction from the hairy root was established to produce natural β‐carotene that achieves the same β‐carotene quantity as that produced by normal roots. This study provides new insight into the production of high‐content and natural β‐carotene for therapeutic application.

Las plantas están constantemente expuestas a diferentes factores bióticos y abióticos, por lo cual deben adaptar su perfil metabólico para defenderse mediante la biosíntesis de metabolitos secundarios. Estos metabolitos aislados o en extracto tienen propiedades aprovechables en el campo farmacéutico, cosmético, agroquímico, alimentario, entre otros. Sin embargo, obtener grandes cantidades de un compuesto natural para estudios que abarquen desde la elucidación estructural, ensayos de diferentes actividades biológicas y hasta su comercialización representa un gran desafío, puesto que, una de las principales limitantes en el estudio de estos metabolitos es la baja cantidad con que se producen en la planta, trayendo como consecuencia un bajo rendimiento en los compuestos purificados, por ello la mayoría de los aportes científicos, si bien representan un avance indiscutible, suele ser información que sólo queda como aporte al conocimiento. Una herramienta biotecnológica utilizada para superar estas limitaciones es el cultivo in vitro de células y tejidos vegetales que, bajo condiciones de elicitación inducen la producción de metabolitos de defensa en las plantas y en consecuencia las enzimas asociadas a su biosíntesis. Piper cumanense es una especie cuyo extracto, aceite esencial y metabolitos aislados han mostrado una prometedora actividad antifúngica, sin embargo, su bajo rendimiento ha impedido realizar investigaciones más profundas a nivel experimental y comercial, lo que ha incentivado la necesidad de buscar una alternativa que permita no sólo la producción de la especie in vitro sino la producción in vitro de sus metabolitos de una forma controlada, permanente y sostenible. Teniendo en cuenta lo anterior, en esta investigación se determinaron los cambios metabólicos en cultivos in vitro de Piper cumanense bajo condiciones de elicitación biótica, y mediante un enfoque enzimático se estimó si estos cambios estuvieron relacionados con las actividades de las enzimas fenilalanina amonio liasa y peroxidasa. Para este propósito, las plántulas in vitro de 5 meses de edad y las suspensiones celulares fueron fortificadas con extracto de micelio acuoso autoclavado de Fusarium solani durante 5 y 15 días para las plántulas in vitro y por 24 y 72 horas para las células en suspensión. Los metabolitos secundarios fueron analizados por cromatografía de gases-espectrometría de masas (GC-MS) y la evaluación de la actividad enzimática de PAL y peroxidasa se realizó espectrofotométricamente. Se encontró que el perfil cromatográfico de los extractos obtenidos de suspensiones celulares y de plántulas in vitro variaban significativamente entre sí. Metabolitos secundarios como fenilpropanoides y terpenos fueron detectados en plántulas in vitro, por el contrario, las suspensiones celulares estaban mayormente compuestas por ácidos grasos. Además, la elicitación en plántulas in vitro durante 5 y 15 días aumentó la producción de apiol y fitol respectivamente, y en ambos casos hubo una sobreexpresión del compuesto No identificado 14 considerado producto directo del proceso de elicitación. Por el contrario, la elicitación de suspensiones celulares durante 24 h no mostró ningún efecto sobre el perfil metabólico, sin embargo, con una exposición de 72 h los fitoesteroles aumentaron significativamente. A nivel enzimático se encontró en plántulas in vitro una inducción significativa de actividad PAL a los 5 días y de peroxidasa a los 15 días de tratamiento, correlacionados con el contenido de miristicina y fitol, respectivamente. En contraste, la elicitación no tuvo efecto sobre ninguna de las dos enzimas en las suspensiones celulares, lo que podría explicar, al menos en parte, la baja producción detectada de metabolitos secundarios. El enfoque utilizado en esta investigación permitió detectar cambios en el perfil metabólico y enzimático de las plántulas in vitro y dejó una evidencia clara que bajo las condiciones de análisis se necesita de cierto grado de diferenciación y madurez celular para que se dé la biosíntesis de metabolitos secundarios.
