Photosynthesis by isolated chloroplasts.

Abstract: Photosynthesis in green plants requires not only photoelectron transport, oxygen evolution, and photophosphorylation, but also the synthetic reactions whereby carbon dioxide is assimilated and reduced to a number of organic compounds via the photosynthetic carbon reduction cycle1 and secondary biosynthetic pathways.2 Complete photosynthesis with isolated chloroplasts has for many years been a goal of biochemists. Photosynthetic reactions could then be isolated from reactions of the cytoplasm, and the cell wall… Show more

“…Young spinach leaves of approximately the same age were harvested from outdoor-grown plants after a 16-hour dark period.-Chloroplasts were -isolated according t9 the method of Jensen and Bassham [11]. The resulting chloroplast suspension in solution ~contained about 2mg chlorophyll/ml.…”

“…For the determination of NADP+ and NADPH levels in chloroplasts in the dark and during co 2 fixation in the light whole choloroplasts were 0 0 -7-isolated and were allo\'/ed to photosynthesize according to the procedure described by Jensen and Bassham [11]. A fluorometric method for measuring pyridine nucleotide levels in animal tissues described by Cartier [15] has been modified to detect very small amounts of pyridine nucleotides in chloroplasts while using a simple and rapid method for preparing samples from a kinetic experiment with ·photosynthesizing chloroplasts.…”

“…After filtration through cheesecloth the suspension was centrifuged for 1 min at 2000 g. The pellet was suspended in the buffer B of Jensen and Bassham [6] . Centrifugation at 30 g (1 min) removed whole cells and greater particles.…”

“…For CO2 -fixation the chloroplasts were incubated in 1 ml of the buffer C used by Jensen and Bassham [6]. The concentrations of NaH14C0s (4.6 mCi/ n-mole) and of chlorophyll were adjusted to those given by the same authors.…”
