Photostability of Contrast Agents for Photoacoustics: The Case of Gold Nanorods

Abstract: Plasmonic particles as gold nanorods have emerged as powerful contrast agents for critical applications as the photoacoustic imaging and photothermal ablation of cancer. However, their unique efficiency of photothermal conversion may turn into a practical disadvantage, and expose them to the risk of overheating and irreversible photodamage. Here, we outline the main ideas behind the technology of photoacoustic imaging and the use of relevant contrast agents, with a main focus on gold nanorods. We delve into th… Show more

“…The plasmonic peaks can be tuned from the visible to the near-infrared (NIR) range depending on their shapes and provide optical contrast, 43 but still has limitations for clinical translation due to photoinstability. 44 Fluorescent proteins have drawn much attention in live-cell optical imaging. 45 GFP, however, has a lower absorption peak wavelength, high fluorescent quantum yield (i.e., low heat conversion efficiency), and shallow imaging depth for in vivo applications.…”

“…(ii) The study of additional excitation wavelengths and multiparametric analysis will allow the opportunity to define additional quantification methods, increasing the classification accuracy further. (iii) Imaging with contrast agents will assess the lymph node metastasis, which is one of the key factors for treatment planning [ 97 , 98 , 99 ].…”

“…It uses the photoacoustic effect as the imaging basis and performs imaging by detecting the ultrasonic waves emitted by biological tissues when absorbing light energy [12]. At the same time of cancer cell metastasis, new blood vessels can be an indicator of early cancer detection, because these new blood vessels contain a large amount of hemoglobin [13], and hemoglobin is the main source of contrast in photoacoustic imaging [14]. This feature makes photoacoustic imaging enjoy great advantages in early cancer detection [15].…”
