Photoinduced Changes in the Chlorophyll a to Chlorophyll b Ratio in Young Bean Plants

Abstract: An alternating light-dark system is described under which etiolated bean (Phaseolus vulgaris) leaves form selectively chlorophyll a.This system consists of cycles of 2 minutes of white light alternating with 98-minute dark periods.Etiolated 80 cm above the plants, were used; one light provided to the plants 120 lux (75-w lamp) and the other, 2000 lux (Phillips Ultrilux, 500 w).The chlorophylls were extracted totally by grinding 1 g of leaves in a mortar with 4 ml of pure acetone, followed by several extractio… Show more

“…Under conditions of continuous illumination the etioplasts quickly show the characteristics of developed chloroplasts (1,13,22). Intermittent illumination (2 min light and 98 min dark), however, transforms the etioplasts into "protochloroplasts" which show higher photochemical activity per Chl than chloroplasts (1), synthesize selectively Chl a, and are devoid of Chl b and grana (3,6), contain "primary" thylakoids and are deficient in the Chl-protein complex II which is considered to be an essential component of grana stacks (4,6).…”

“…6-day etiolated Phaseolus vulgaris leaves were used, grown and handled as in [1]. The leaves with one cotyledon removed were either exposed to ImL [2 rain white light alternating with 98 rain dark in cycles], or to CL for 18 h and then transferred to the dark.…”

“…Leaves handled in the same manner but immersed in H20 were used as controls. Chl was extracted from leaves as in [1] and determined according to [13]. PS I and PS II activities were monitored as in [ 11 ].…”
