Photodynamic Effect of Methylene Blue and Low Level Laser Radiation in Head and Neck Squamous Cell Carcinoma Cell Lines

Abstract: Photodynamic therapy (PDT) is suggested to have an impact on the treatment of early stage head and neck cancers (HNSCC). We investigated the effect of PDT with methylene blue (MB) and a diode laser (660 nm) as the laser source on HNSCC cell lines as an in vitro model of surface oral squamous cell carcinoma. Cell-cultures were exposed to 160 µM MB for 4 min and to laser light for 8 min. Viability was proven via cell viability assay and clonogenic survival via clone counting assay. The combination of MB and diod… Show more

“…One-thousand cells from all samples were plated in 75-cm 2 cell culture flasks grown in serum-supplemented medium for 3 weeks and the growing colonies were stained with gentian violet and were counted. The number of growing colonies were related to plated cell numbers (1000) [57].…”

“…Mouse Gardner lymphoma, sarcoma 180, Ehrlich ascites, and mammary adenocarcinoma have all been successfully treated using MB in animal models [82]. Likewise, some human cancer cells have been reported as responders of MB-mediated light therapy, including HeLa cervical adenocarcinoma cells, early stage head and neck squamous cell carcinoma (HNSCC), and lung adenocarcinoma cells [83][84][85]. Although some studies have indicated that the cytotoxicity of MB can be improved by chemical modifications, these studies are beyond the scope of this review, particularly considering the functionality of the safe, proven molecules in the treatment of a variety of cancers using MB-mediated light therapy.…”

“…To quantify the relative photoactivity of MPPa and NMPi in the absence of tumor cells, 1 O 2 photogeneration was measured by 1,3-diphenylisobenzofuran (DPBF) as a selective 1 O 2 acceptor ( Figure 5 b). NMPi presented slightly better 1 O 2 photogeneration than MPPa, and NMPi was almost comparable to methylene blue (MB) as a standard 1 O 2 sensitizer [ 39 ]. This result was consistent with the cell viability results shown in Figure 2 and Figure 3 .…”
