Photoacoustic imaging of early inflammatory response using gold nanorods

Abstract: Gold nanorods have unusually strong absorption in near infrared, which can be utilized for an optical imaging with nanocolloids. The feasibility of photoacoustic imaging of inflammatory responses using bioconjugated gold nanorods is demonstrated. To target the stimulated cells, gold nanorods were conjugated to anti-intercellular adhesion molecule-1 (ICAM-1) which binds to cell surfaces over expressing ICAM-1. A monolayer of stimulated endothelial cells labeled with bioconjugated gold nanorods was scanned using… Show more

“…S3). It has been demon- strated that a variety of gold nanocolloids are already entering in vivo clinical trials (19,(29)(30)(31)(32)(33). Among them, Au nanorods present particularly good optical absorption in the near-infrared region, tunable by changing the aspect ratio.…”

“…11 Absorption of light by GNRs is followed by predominantly nonradiative de-excitation processes and the released heat and subsequent temperature rise have applications in improving photoacoustic signals. 12,13 Further the temperature rise can be made sufficiently high to cause cell death by hyperthermia, which has potential for important therapeutic applications. [14][15][16] In the last mentioned application, the ability to redshift LP peaks by tailoring the dimensions of the GNRs coupled with the fact that biological tissue is relatively transparent in the red and NIR wavelengths makes GNRs very attractive.…”

“…Excess PAA in solution was removed by brief centrifugation and GNRs were dispersed in 10 ml of PBS followed by the addition of 1.25 ml of 1M N-ethyl-N-3-dimethylamino propyl-carbodiimide (EDC) (Sigma-Aldrich, St. Louis, MO, USA) and 1.25 ml of 1M N-hydroxy-succinimide (NHS) (Sigma-Aldrich, St. Louis, MO, USA). [10][11][12][13][14]16] After 20 min, the reaction mixture was added to 20 µl of FITC labeled anti-ICAM-1 antibody or PE labeled anti-E-selectin antibody. The EDC/NHS mixture forms an active ester intermediate with the -COOH groups of PAA which undergoes amidation reaction with the -NH2 group in the FITC labeled anti-ICAM-1 and PE labeled anti-E-selectin antibodies to yield the GNR-antibody conjugates.…”
