Phosphorylation-dependent power output of transgenic flies: an integrated study

Abstract: We examine how the structure and function of indirect flight muscle (IFM) and the entire flight system of Drosophila melanogaster are affected by phosphorylation of the myosin regulatory light chain (MLC2). This integrated study uses site-directed mutagenesis to examine the relationship between removal of the myosin light chain kinase (MLCK) phosphorylation site, in vivo function of the flight system (flight tests, wing kinematics, metabolism, power output), isolated IFM fiber mechanics, MLC2 isoform pattern, … Show more

“…Aluminum T-clips were attached to each segment, yielding a preparation Ϸ0.6 mm long and Ϸ0.1 mm wide. The segment was transferred to a 30-l drop of glycerol-and detergent-free relaxing solution and mounted between a piezoelectric motor and strain gauge on a mechanical rig (25). The temperature was set at 15°C.…”

“…For each solution condition, the complex modulus of each fiber was fitted to a three-term equation (25,30) by following the Predicted shifts of apparent rate constants from calculations based on eight schemes with different rate-limiting steps (corresponding to numbers in the cross-bridge scheme in C). The details of the calculations are given in supporting information.…”

“…Fibers were immediately used for mechanical experiments or were transferred to storage solution (relaxing solution plus protease inhibitor mixture and 50% glycerol) at Ϫ20°C and used within a few days of dissection. Skinned fibers, in relaxing solution, were attached to aluminum T-clips at both ends and mounted to a force transducer and a length driver (23,41). The fiber was stretched until just taut (minimal stress), and the initial fiber length was measured.…”

“…To perform sinusoidal analysis, small-amplitude sinusoidal length changes (0.125% muscle length) were applied to the fiber at 47 frequencies (0.5-1,000 Hz) while measuring the force response (23). Length and force were normalized to determine strain (⌬L͞L) and tension (F͞CSA) by dividing the length change (⌬L) by total fiber length (L) and by dividing the force (F) by the fiber crosssectional area (CSA).…”
