Phospholipase Dα6 and phosphatidic acid regulate gibberellin signaling in rice

Abstract: Phospholipase D (PLD) hydrolyzes membrane lipids to produce phosphatidic acid (PA), a lipid mediator involved in various cellular and physiological processes. Here, we show that PLDa6 and PA regulate the distribution of GIBBERELLIN (GA)-INSENSITIVE DWARF1 (GID1), a soluble gibberellin receptor in rice. PLDa6knockout (KO) plants display less sensitivity to GA than WT, and PA restores the mutant to a normal GA response. PA binds to GID1, as documented by liposome binding, fat immunoblotting, and surface plasmon … Show more

“…For At AHL4, PA-mediated tethering to the nuclear membrane was excluded as a mechanism of PA action, suggesting a hypothetical possible association of PA with At AHL4 beyond membranes [ 3 ]. PA also binds to the gibberellin receptor Os GID1, promotes its nuclear localization, and mediates the gibberellin response in rice [ 36 ].…”

“…In rice protoplasts, gibberellin induces OsPLDα6 translocation from cytosol to nuclei. PA formed by OsPLDα6 activity positively mediates gibberellin signaling via PA binding to gibberellin receptor OsGID1, promotion of its nuclear localization and degradation of the DELLA protein SLENDER RICE1, a negative regulator of the hormone signaling [ 36 ]. In contrast, the level of suppressor protein SLENDER RICE1 was decreased in npc6 rice mutants treated with gibberellin, but drastically increased in OsNPC6 over-expressing plants [ 122 ].…”

“…In Arabidopsis, PA mediates the tethering of ABI1 (a negative regulator of ABA) to the plasma membrane to prevent it from reaching its downstream targets in the nucleus [ 29 ]. PA could bind to GA receptor OsGID1 to promote its nuclear localization for gibberellin response in rice [ 30 ]. PA is also involved in the process of ETH signaling by binding to CTRI, the negative regulator in the ethylene signaling pathway, and inhibiting its activity.…”

“…A small change in PA level may result in a substantial effect due to its function in the upstream of signaling cascades [ 20 ]. Phosphatidic acid with various species of acyl group in specific membrane region has been shown to bind to various effectors involved in diverse cellular processes in plants [ 19 , 20 , 21 ]. In plants, PA can be generated through three major routes [ 22 ].…”
