Phosphoglycerate mutase, 2,3-bisphosphoglycerate phosphatase and creatine kinase activity and isoenzymes in human brain tumours

Abstract: Summary The distribution of phosphoglycerate mutase (EC 5.4.2.1, PGM), 2,3-bisphosphoglycerate phosphatase (EC 3.1.3.13, BPGP) and creatine kinase (EC 2.7.3.2, CK) activity and isoenzymes in various regions of adult human brain and in brain tumours (astrocytomas, anaplastic astrocytomas, glioblastomas and meningiomas) has been determined using electrophoresis. PGM and cytosolic CK exist in mammalian tissues as three isoenzymes that result from the homodimeric and heterodimeric combinations of two subunits [typ… Show more

“…Tumor formation is correlated with a reduction of the 2,3-BPG-synthesizing enzyme 2,3-bisphosphoglycerate mutase (E.C. 5.4.2.4) (37)(38)(39)(40)(41). Because tumor cells are characterized by high glycolytic rates although 2,3-BPG is not detectable, another 2,3-BPG-independent PGM-activating mechanism can be postulated.…”

“…28,29 Brain tumours possessed lower PGM activity than normal brain. 30 In fibroblasts, exposure of the cells to hypoxia induces induction of PGM activity, concomitant with elevations of mRNA and the protein levels of PGM-B, indicating that the enzyme activity is regulated mainly at the transcriptional step. 31 Thus, the colorectal cancer specimens analyzed in our study were expected to have high PGM-B contents.…”

“…During glycolysis, PGM catalyzes the interconversion of 3-phosphoglycerate to 2-phosphoglycerate, and then α-enolase catalyzes the conversion of 2-phosphoglycerate to phosphoenolpyruvate [28]. PGM1 is one of the PGM isozymes, and α-enolase is a subunit of enolase.…”
