Phosphatidylinositol 5-phosphate 4-kinase regulates early endosomal dynamics during clathrin-mediated endocytosis

Abstract: Endocytic turnover is essential for the regulation of the protein composition and function of the plasma membrane, and thus affects the plasma membrane levels of many receptors. In Drosophila melanogaster photoreceptors, photon absorption by the G-protein-coupled receptor (GPCR) rhodopsin 1 (Rh1; also known as NinaE) triggers its endocytosis through clathrin-mediated endocytosis (CME). We find that CME of Rh1 is regulated by phosphatidylinositol 5 phosphate 4-kinase (PIP4K). Flies lacking PIP4K show mislocaliz… Show more

“…Prior studies have demonstrated a role for PI5P4Ka and PI5P4Kb in clathrin-mediated transport and autophagy (Kamalesh et al, 2017;Lundquist et al, 2018), highlighting a possible vesicular distribution for the PI5P4Ks. To investigate their subcellular localization, we performed double immunofluorescence of PI5P4Ka and PI5P4Kb with a panel of subcellular markers.…”

“…These kinases also regulate cellular growth pathways, including the mammalian target of rapamycin (mTOR) and phosphatidylinositol 3-kinase (PI3K) pathways (Carricaburu et al, 2003;Gupta et al, 2013). Furthermore, they regulate early endosomal homeostasis during clathrin-mediated endocytosis, a key step in synaptic vesicle trafficking (Kamalesh et al, 2017). Of particular interest, these enzymes also play a major role in suppressing the activity of the type 1 phosphatidylinositol-4-phosphate 5-kinases through direct binding to these enzymes (Wang et al, 2019).…”

“…A recent study indicated that in Drosophila photoreceptors, dPIP4K regulates 145 clathrin-mediated endocytosis (CME) from the plasma membrane, and its absence 146 causes increased CME uptake, leading to an increased number of Rh1-loaded early 147 endocytic vesicles (Kamalesh et al, 2017). This cytoplasmic accumulation of Rh1 in 148 dPIP4K-deficient photoreceptors is light-dependent, likely because of light-stimulated 149 endocytosis, and this phenotype is rescued by the inhibition of endocytosis by 150 Rab5RNAi.…”
