Phorbol myristate acetate-induced modulation of antibody-dependent cellular cytotoxicity by human polymorphonuclear leukocytes

Abstract: The effect of phorbol myristate acetate (PMA) on the antibody-dependent cellular cytotoxicity (ADCC) of human polymorphonuclear leukocytes (PMNs) against human erythroleukemic K562 cells was studied by the use of a 3-h 51Cr-release assay. Pretreatment of PMNs with PMA (10 ng/ml) for 60 min resulted in inhibition of subsequent ADCC. This inhibition was dependent on doses of PMA. The effect of pretreatment of PMNs with PMA on O2- generation of the cells was also studied. The ability of the cells to generate O2- … Show more

“…Many of the reports listed in Tables I and II discuss the anticancer effect of PMN via ADCC,27, 41, 46, 50, 64, 83, 116, 120, 122–126, 128, 129, 131–177 and many provide direct evidence of this antibody‐mediated antitumoral role of PMN in vitro. The first reports were published in 1974 and 1975 by Gale and Zighelboim 131, 132.…”

“…Effect of pretreatment with low concentrations of PMA on 02 generation in the absence of extracellular Ca2 . PMA is known not only to be a potent biological stimulator but also to have a potentiating effect on cellular function (9,19). To examine whether Ca2+-dependent O,-generation by stimulation with YI 51 is modulated by pretreatment with low concentrations of PMA, the amount of 02-generation induced by YI 51-bound PMN preincubated with the reagent over a concentration range of 1010 to 10-9 M in Ca2+-free medium for 10 min was measured after addition of F(ab')2 fragments of rabbit anti-mouse immunoglobulin antibody.…”

“…We did, however, determine that YI 51 appears to bind to a molecule other than Fc,rR or to an epitope on Fc,rR that is distinct from the Fc-binding site because of the following findings. (i) YI 51 could not bind to cells of HL 60 and K 562 which had been reported to have Fc,R ( PMA is known not only to be a potent biological stimuilus but also to have a potentiating effect on cell function (9,19). We thus investigated whether pretreatment of PMN with low concentrations of PMA was able to modulate O-generation by the cells subsequent to stimulation.…”
