Phenotypic Variation ofHelicobacter pyloriIsolates from Geographically Distinct Regions Detected by Lectin Typing

Abstract: A total of 309 Helicobacter pylori isolates from 18 different countries were analyzed with a previously developed lectin typing system. The system was developed by using a proteolytic pretreatment to enhance the carbohydrate fraction of the sample. Four lectins from Ulex europaeus, Lotus tetragonolobus, Erythrina cristigali, and Triticum vulgaris were used to type the strains. The lectins were chosen for their specificities for sugars commonly encountered in the lipopolysaccharide of H. pylori. The isolates we… Show more

“…Consistent with previous studies by others (Hynes et al , 1999, 2002), the lipopolysaccharide lectin‐typing patterns in our clinical isolates were highly variable. This was also the case for the mouse‐adapted strains, with no lectin‐typing pattern being associated with the ability of H. pylori strains to infect mice.…”

“…Preparation of bacterial lipopolysaccharides was as described previously by others (Hynes et al, 2002). Briefly, H. pylori samples were resuspended in 5 mL sterile PBS and disrupted by sonication on ice (Branson Sonifier Cell Disruptor) by 30-s pulses 10 times, with 1 min between each pulse.…”

“…The ability of carbohydrate binding proteins to interact preferentially with specific cell types has also led to attempts to use them as drug carrier (Sharma et al ., 2004) and for blood typing (Khan et al ., 2002). Important further applications include the diagnostic use of lectins for the identification of microorganisms (Hynes et al ., 2002) and the use of lectins for the visualization of extracellular matrices to determine the carbohydrate composition of, e.g. biofilms (Neu et al ., 2001).…”

“…Isolates of H. pylori, irrespective of geographical location, have identical phenotypes as defined by conventional bacteriological tests (Owen et al, 2001b) although there is evidence that strains can differ in some expressed features such as vacuolating cytotoxin activity (Forsyth et al, 1998), lectin reaction patterns (Hynes et al, 2002) and interleukin-8 induction in gastric epithelial cells (Owen et al, 2003). In contrast, strains of H. pylori from unrelated individuals exhibit an unusually high level of genomic diversity as indicated by various fingerprinting techniques (Owen et al, 2001b).…”
