Phenotypic heterogeneity of spontaneous lymphomas of CWD mice

Abstract: Animals of the inbred mouse strain, CWD, express endogenous murine leukemia viruses early in life and have a high incidence of spontaneous neoplasms. We found that approximately one half of these animals died of malignant lymphoma by the age of 16 months. Splenic enlargement was seen in all mice, but thymic involvement was unusual. One half of the CWD tumors were diffuse lymphoblastic or immunoblastic lymphomas while the remainder were large cell, small cell, or mixed cell lymphomas. Analysis of DNAs from 12 t… Show more

“…T-cell tumors were defined by detectable TCRJ3 rearrangements in the DNA in the absence of JK rearrangements and if the cells stained with Thy 1.2 antibody of tissue sections. JH rearrangements were seen in some of the T-cell lymphomas as previously reported (19,51).…”

“…Extraction of genomic DNA and Southern blotting of the tumor DNAs were performed as described previously (35). The immunophenotype of the tumors was based on histopathologic testing, the presence or absence of immunoglobulin and T-cell receptor gene rearrangements, and immunocytochemical analysis by techniques detailed in an earlier report (49). The probes for gene rearrangement studies included portions of the J regions of the immunoglobulin heavy chain (JH) and kappa light chain (JK) and the constant region of the murine T-cell receptor beta chain.…”

“…Ten HRS/J and 14 CWD mice were monitored until death or were sacrificed because of advanced disease. The presence of lymphoma was confirmed by necropsy and histologic examination of tumor tissues (33).…”
