Phenotypic and genotypic analysis of Borrelia burgdorferi isolates from various sources

Abstract: A total of 17 B. burgdorfieri isolates from various sources were characterized by sodium dodecyl sulfate-polyacrylamide gel electrophoresis of whole-cell proteins, restriction enzyme analysis, Southern hybridization with probes complementary to unique regions of evolutionarily conserved genes (16S rRNA and fla), and direct sequencing of in vitro polymerase chain reaction-amplified fragments of the 16S rRNA gene. Three groups were distinguished on the basis of phenotypic and genotypic traits, the latter traced … Show more

“…Strains of B. burgdorferi sensu lato related to Lyme disease are highly heterogeneous in their gene structures, and the three genospecies, B. burgdorferi sensu stricto, B. garinii and B. afzelii, have been proposed on the bases of DNA homology (3,9,28), and 16S rRNA gene and fla restriction analyses (1). Japanese isolates were characterized bacteriologically based on protein profiles (25), plasmid profiles (32), restriction fragment length polymorphism (RFLP) analysis in 23S rRNA gene, 16S rRNA gene (12,13,15,19) and fla (19) and reactivities with MAbs against Osp A and flagellin (25).…”

“…Genetic, biochemical and immunological studies revealed significant differences among the Lyme disease borreliae isolated from various biological and geographical sources (1,25,27,28,40). Previous reports (13)(14)(15), and our recent studies on the basis of the reactivities with 16 monoclonal antibodies to outer surface protein A and DNA-DNA hybridization indicated that B. garinii strains isolated in Japan are heterogeneous and different from B. garinii isolates in Europe (33).…”

“…To date, the causative agent of the disease has been found to consist of at least three genospecies that are human pathogens; Borrelia burgdorferi sensu stricto (s.s.), Borrelia garinii and Borrelia afzelii. Some differences in disease manifestation are associated with infection by each of these genospecies (1,2,8).…”

“…A number of Borrelia isolates have been obtained from diverse geographic locations. Many Borrelia isolates have been examined by monoclonal antibody (1,4,5), plasmid profile analysis (6,17,18,30), and DNA restriction pattern analysis (18,22). Recently, on the basis of DNA homology, specific rRNA gene restriction patterns, and protein electrophoresis patterns, three genospecies for the strains of B. burgdorferi were proposed (2).…”
