Phenotype Identification of HeLa Cells Knockout CDK6 Gene Based on Label-Free Raman Imaging

Abstract: Identifying cell phenotypes is essential for understanding the function of biological macromolecules and molecular biology. We developed a noninvasive, label-free, single-cell Raman imaging analysis platform to distinguish between the cell phenotypes of the HeLa cell wild type (WT) and cyclin-dependent kinase 6 (CDK6) gene knockout (KO) type. Via large-scale Raman spectral and imaging analysis, two phenotypes of the HeLa cells were distinguished by their intrinsic biochemical profiles. A significant difference… Show more

“…Further semiquantitative analysis by integrating the intensities of individual Raman bands revealed significantly different peaks predominantly located at 749 and 1128 cm −1 (cytochrome C), 854, 925, and 1659 cm −1 (collagen), 1003 cm −1 (phenylalanine), 1083 cm −1 (P-O stretching vibrations; DNA/phospholipids), 1309 and 1449 cm −1 (lipids), and 1365 and 1586 cm −1 (vitamin A) (Figure 1D). [27][28][29][30][31][32][33] It showed that with the prolongation of CCl 4 dosing in mice, the Raman peaks assigned to collagen progressively intensified, which echoed the foregoing biochemical results, mirroring the cascading exacerbation of fibrotic injury. Analogous incremental enhancement was ascribed to the Raman peaks of lipids and cytochrome C, suggesting potential lipid accumulation and mitochondrial dysfunction in fibrotic livers.…”

“…4e, the experimental parameters can be found in the gure caption). Raman images of two bands at the ngerprint region (1445 cm − 1 : CH 2 and CH 3 deformation of lipids and proteins, 1656 cm − 1 : C = C stretching of lipids), and two bands at the CH abundant region (2867 cm − 1 : CH 2 symmetrical stretching of lipids, 2915cm − 1 : CH band of lipids and proteins) 48,49 are presented. The raw images (Fig.…”

“…The required complex instrumentation also requires careful beam synchronization. A number of compound microscopes that combine Raman imaging with traditional optical microscopes have demonstrated added value to that of Raman imaging alone (72)(73)(74)(75)(76)(77)(78). These compound microscopes incorporate Raman imaging with bright-field, fluorescence, or confocal microscopy to provide simultaneous information on lipids and other cellular constituents such as proteins, including their relative spatial locations (75,(79)(80)(81)(82)(83)(84).…”
