Phenolic Constituents from Wendlandia tinctoria var. grandis (Roxb.) DC. Stem Deciphering Pharmacological Potentials against Oxidation, Hyperglycemia, and Diarrhea: Phyto-Pharmacological and Computational Approaches

Abstract: Wendlandia tinctoria var. grandis (Roxb.) DC. (Family: Rubiaceae) is a semi-evergreen shrub distributed over tropical and subtropical Asia. The present research intended to explore the pharmacological potential of the stem extract of W. tinctoria, focusing on the antioxidant, hypoglycemic, and antidiarrheal properties, and to isolate various secondary metabolites as mediators of such activities. A total of eight phenolic compounds were isolated from the dichloromethane soluble fraction of the stem extract of t… Show more

“…By blocking the enzymes α-amylase, α-glucosidase, and human aldose reductase, flavonoid glycosides have shown to have anti-diabetic activities in several prior investigations [ 5 , 57 59 ]. Due to the rarity of these two isolated flavonoid glucosides (compounds 1 and 2) and the lack of available experimental data, it is possible to infer their specific mode of action against diabetes from the actions of their analogues.…”

“…These plant extracts rich in flavonoids may be an effective treatment for both acute and chronic diarrhea because of their capacity to reduce intestinal fluid output and slow bowel motions. Additionally, they might protect the intestines from oxidative stress and preserve the mucosal lining's integrity, potentially minimizing the severity of long‐term inflammatory damage 71 …”

“…Additionally, they might protect the intestines from oxidative stress and preserve the mucosal lining's integrity, potentially minimizing the severity of long-term inflammatory damage. 71 Preclinical studies in developing new drugs must include toxicity assessments in suitable animal models. Due to people's unwavering belief in the efficacy of the herb and extensive traditional/folk usage of plants, toxicological testing of natural phytoconstituents, medicinal plants, or any new medicine for their harmful potential becomes essential.…”

“…The anti-diarrheal activity of SSCFE was evaluated using the method of castor oil induced diarrhea in mice (Jannat et al, 2022;Farzana et al, 2022). According to this method, each mouse was fed with 1ml of highly pure analytical grade of castor oil which would induce diarrhea.…”

“…This activity is an indication of its use in various stomach associated disorders like cramps, constipation, indigestions etc. One of the various mechanisms by which castor oil induces diarrhea is electrolyte permeability via endogenous prostaglandin secretion (Anjum et al, 2022;Farzana et al, 2022;Jannat et al, 2022). One of the potential reasons by which the SSCF shows its antidiarrheal activity by inhibiting prostaglandin synthesis in the intestine is evident by the fact that the feces after treatment were rather dry (Bahekar and Kale, 2015).…”
