Stevia is a plant with many beneficial properties. It contains not only steviol glycosides, which are used as non-caloric natural sweeteners, but also a number of metabolites with antioxidant properties. This study examined the content of both phenolic acids and flavonoids in stevia leaves as an effect of treating seeds with melatonin and conducting germination in NaCl conditions. The results of our research indicated higher amounts of phenolic acids compared to flavonoids in stevia leaves. Among these acids, isochlorogenic, rosmarinic, and chlorogenic acids were accumulated in the largest amounts, regardless of the germination conditions. For 5 and 100 µM of melatonin treatments, the content of both phenolic acids and flavonoids increased. However, in salinity conditions (50 mM NaCl), 500 µM of melatonin had the most favorable effect on the synthesis of phenolic acids. The phenolic acids in that case reached a level three-times higher than that in the samples with the same melatonin concentration but without NaCl. We also found that the content of phenolic compounds varied depending on the age of the leaves. To the best of our knowledge, this is the first study to describe the effect of melatonin and NaCl on the synthesis on phenolic acids and flavonoids in stevia.

Stevia rebaudiana Bertoni possesses various medicinal and food industrial applications. This study is the first to explore the effect of the cytokinins meta-Topolin (mT; 6-(3-hydroxybenzylamino) purine), zeatin, kinetin, and BAP (6-benzylaminopurine) at concentrations of 0 (control), 5, 10, and 15 µM on shoot multiplication, as well as stevioside, rebaudioside A, phenolic acid, and flavonoid content in bioreactor cultures. The highest number of shoots (23.4 per explant) was obtained in the medium containing 5 μM of mT. However, 15 μM of mT was superior for fresh biomass production and dry biomass accumulation. Reversed-phase (RP)-HPLC analysis showed a beneficial effect of 5 μM mT on stevioside (11.43 mg/g dry weight [DW]) and rebaudioside A (10.74 mg/g DW) biosynthesis. In all conditions, the ratio of rebaudioside A/stevioside ranged from 0.75 to 1.12. The phenolic acids chlorogenic, neochlorogenic, isochlorogenic A, and rosmarinic were confirmed in the stevia extracts, as were the flavonoids isoquercetin, and quercitrin. The highest accumulations of chlorogenic and neochlorogenic acids and flavonoids were observed in shoot tissues derived from 5 µM mT, whereas 5 µM of BAP stimulated biosynthesis of chlorogenic, isochlorogenic A, and rosmarinic acids. This is the first report on the use of mT-cytokinin showing high potential in stevia cultures.

Introduction: It is important to counteract the oxidative stress and inflammatory burden occurring in the body. The use of herbal components obtained via green synthesis, such as Stevia and peppermint which are alcohol-free and comprise relatively of less chemical components and have been therapeutically found effective for antidiabetic properties can prove to be more efficient and biocompatible to tackle the same. Aim: The aim of the study was to evaluate the anti-inflammatory and antioxidant properties of a herbal formulation of stevia rebaudiana and peppermint at varying concentrations. Materials and Methods: The study was performed as an in vitro study under a laboratory setting. Synthesis of the herbal formulation was performed using stevia and peppermint in the lab. Subsequently the formulation was tested for its anti-inflammatory activity using the protein denaturation assay and antioxidant activity using the DPPH Assay method at various concentrations. The obtained values were compared with that of the known standard. The statistical analysis was done using IBM SPSS software analysis version 23. Results: Anti inflammatory property of the Steviaa and peppermint herbal formulation was significantly less at 10μL concentration but at higher concentrations it was comparable to the standard. Regaling the antioxidant property, the herbal formulation showed a concentration dependent increase from lower concentration to higher concentration but at all levels it was significantly lesser than the standard. (p<0.05) Conclusion: The herbal formulation of Stevia and peppermint is a potent anti-inflammatory agent. It possesses antioxidant properties, however which are not as effective as the standard.
