We present a multiple-path scalable configuration that allows the interrogation of an intralipid flow in a vessel at five multiple depths separated by a spatial interval that could be easily adjusted from zero to values less, larger, or much larger than the coherence length of the optical source. The range of flow values measured is accommodated by modifying the difference between the frequencies of signals driving two frequency shifters, each being a part of an active loop placed in each arm of a Mach-Zehnder interferometer.

Ciliary motion plays a critical role in the overall respiratory health of the upper airway. These cilia beat at a native frequency and in a synchronized pattern to continuously transport foreign particulate trapped in a layer of mucous out of the upper airway. Disruption of ciliary motion can lead to severe respiratory diseases and compromised respiratory function. Currently, the study of cilia requires expensive high speed cameras and high powered microscopes which is unsuitable for in vivo imaging and diagnosis. Doppler based optical coherence tomography has the potential to visualize the microscopic motion of cilia during their beating cycle. We demonstrate the development of a high-speed Doppler optical coherence tomography system that not only can rapidly determine the cilia beat frequency, but also simultaneously visualize the temporal cilia beating pattern which plays critical roles in cilia function.

Optical Coherence Tomography (OCT) was originally conceived as a volumetric imaging method. Quickly, OCT images went beyond structural data and started to provide functional information about an object enabling for example visualization of blood flow or tissue elasticity. Minimal or no need for system alterations make functional OCT techniques useful in performing multimodal imaging, where differently contrasted images are produced in a single examination. We propose a method that further extends the current capabilities of OCT and requires no modifications to the system. Our algorithm provides information about the sample’s Group Velocity Dispersion (GVD) and can be easily applied to any OCT dataset acquired with a Fourier domain system. GVD is calculated from the difference in material’s optical thickness measured from two images obtained for different spectral ranges. Instead of using two separate light sources, we propose to apply a filter-based, numerical procedure that synthesizes two spectra from one broadband spectrum. We discuss the limitations of the method and present GVD values for BK7 and sapphire and ocular media: cornea and aqueous humour of a rat eye. Results corroborate previous measurements using two different light sources.
