Phase and Index of Refraction Imaging by Hyperspectral Reflectance Confocal Microscopy

Abstract: A hyperspectral reflectance confocal microscope (HSCM) was realized by CNR-ISC (Consiglio Nazionale delle Ricerche-Istituto dei Sistemi Complessi) a few years ago. The instrument and data have been already presented and discussed. The main activity of this HSCM has been within biology, and reflectance data have shown good matching between spectral signatures and the nature or evolution on many types of cells. Such a relationship has been demonstrated mainly with statistical tools like Principal Component Analy… Show more

“…Conversely from other literature works that have tackled macrophagic polarization using spectroscopic tools addressing specific light-matter interactions ( e . g ., fluorescence lifetime imaging microscopy 26 , 27 , forward angular light scattering 28 ) for the establishment of decision support systems based on chemometry, the proposed approach exploits a simpler physical phenomenon, namely Vis-NIR reflectance 54 , for the analysis of cells in vitro . As a major advantage, our system has the potential for easy integration into routine equipment (microscopes), or even into cytometric setups, and is in principle compatible with other wide-spectrum light sources, such as cost-effective metal halide lamps.…”

“…(34,35) The capability to separate light scattered from different focal planes is of significance to the design of our microscope because this light is responsible for undesired spectral blurring and prevents the collection of reflectance spectra at the highest possible spatial resolutions. (36)(37)(38)(39) Surface Shape Estimation and Min-Max Projections. Although careful sample preparation can produce samples with mirror-like finishes to the unaided eye, based on the resolution of our microscope, even techniques like microtomy or mechanical polishing with fine diamond slurries will leave cross sections with surface microfacets that can introduce angle dependent highlights and shadow artifacts.…”
