Pharmacovigilance Signals of the Opioid Epidemic over 10 Years: Data Mining Methods in the Analysis of Pharmacovigilance Datasets Collecting Adverse Drug Reactions (ADRs) Reported to EudraVigilance (EV) and the FDA Adverse Event Reporting System (FAERS)

Abstract: In the past twenty years, the consumption of opioid medications has reached significant proportions, leading to a rise in drug misuse and abuse and increased opioid dependence and related fatalities. Thus, the purpose of this study was to determine whether there are pharmacovigilance signals of abuse, misuse, and dependence and their nature for the following prescription opioids: codeine, dihydrocodeine, fentanyl, oxycodone, pentazocine, and tramadol. Both the pharmacovigilance datasets EudraVigilance (EV) and… Show more

“…However, a recent study evaluating the pharmacovigilance misuse/abuse signals related to semaglutide from an Food and Drug Administration Adverse Events Reporting System dataset found significantly greater misuse associated with semaglutide as compared with other GLP-1 RAs. The same comparison of semaglutide with the phentermine-topiramate combination was not associated with any significant differences 4 …”

“…The same comparison of semaglutide with the phenterminetopiramate combination was not associated with any significant differences. 4 Data on abusing drugs potentiating weight loss are also sparse in classic AN. One report described a 50-year-old woman with no history of diabetes, who presented to the emergency department with a BMI of 11 kg/m 2 , confusion, severe abdominal pain, and renal failure after misusing metformin obtained over the Internet for weight loss.…”

“…The rise in the misuse of opiates/opioids is becoming a health challenge globally. Opiate/opioids’ misuse is an overlooked and concerning phenomenon which most typically has an iatrogenic origin, as it begins with unnecessary/inappropriate drug prescription [ 30 ]. Indeed, these molecules may impair male spermatogenesis via different pathways; androgen suppression with associated hypogonadism may be associated with a direct gonadotropin release inhibition and/or with a hypothalamic–pituitary–gonadal axis impairment secondary to opioid-induced prolactin release.…”

“…We used the previously selected preferred terms. The program is presenting an ADR as putative whenever the followings are true: report count >3, PRR >2, and χ2 >4 ( Böhm et al, 2021 ; Chiappini et al, 2022 ).…”
