Lead Induced Hepato-renal Damage in Male Albino Rats and Effects of Activated Charcoal

Lead is a multi-organ toxicant implicated in various cancers, diseases of the hepatic, renal, and reproductive systems etc. In search of cheap and readily available antidote this study has investigated the role of activated charcoal in chronic lead exposure in albino rats. Eighteen mature male albino rats were used, divided into three groups of six rats per group. Group 1 (control rats) received deionised water (10 ml/kg), group 2 was given lead acetate solution 60 mg/kg and group 3 rats were given lead acetate (60 mg/kg) followed by Activated charcoal, AC (1000 mg/kg) by oral gavage daily for 28 days. Rats in group 2 showed significant increases in serum Aspartate aminotransferase, Alkaline phosphatase, Alanine aminotransferase, urea, bilirubin, total cholesterol, triglycerides, Low Density Lipoprotein, Very Low Density Lipoproteins, Total White Blood Cell Counts, Malondialdehyde, Interleukin-6, and decreases in Packed Cell Volume, hemoglobin concentration, Red blood cell count, total proteins, albumins, superoxide dismutase, glutathione peroxidase and total glutathione. Co-administration of AC significantly decreased these biomarkers with the exception of the sperm parameters. Histopathology of liver and kidney also confirmed the protective effective of AC against lead induced hepato-renal damage. AC may be beneficial in chronic lead induced liver and kidney damage.

Aim:The present study aimed to investigate the antioxidant and hepatoprotective activity of an extract of Sphaeranthus indicus Linn leaves against the carbon tetrachloride (CCl 4 )-induced hepatotoxicity using in vitro and in vivo models of liver injury. Materials and Methods: The coarsely powdered leaves of S. indicus Linn were successfully extracted with petroleum ether extract (SIPEE), chloroform extract (SICHE), ethyl acetate extract (SIEAE), ethanolic extract (SIEE), and aqueous extract (SIAQE). The total tannin content, total phenolic content, and total flavonoids content were evaluated. Extracts were administered to evaluate in vitro human live hepatoma cell line study and in vivo hepatoprotective activity against CCl 4 intoxicated mice. Results: Different extracts of SI presented reducing power in the order of Vitamin C >SIEE >SIEAE >SICHE >SIAQE >SIPEE. At the concentration 200 μg/mL, SIEE showed 50.4% 2,2-diphenyl-1-picrylhydrazyl scavenging property, 57.1% hydroxyl radical° scavenging activity, 46.1% nitric oxide° (NO°) radical scavenging property, and 73.2% superoxide radical scavenging action. The treatment with SIEE prevented the hepatic malondialdehyde level by 22.22, 26.67, and 58.89% with the doses of 100, 200, and 400 mg/kg, respectively. Conclusion: This study suggested that the presence of flavonoids in S. indicus Linn leaves exhibiting marked antioxidant and hepatoprotective activities.

Introduction: This work was done to determine phytochemical content, antioxidant activity, and hepatoprotective effects of Zizyphus xylopyrus leaves extracts against carbon tetrachloride-induced hepatotoxicity in in vitro and in vivo models. Materials and Methods: The total flavonoids content (TFC), total phenolic content (TPC), and total tannin content (TTC) were determined using quercetin and tannic acid equivalents, as standard while antioxidant activities of extracts were determined using the standard in vitro methods. All the extracts subjected to in-vitro HepG2 cell line study as well as to evaluate in vivo hepatoprotective effects against carbon tetrachloride (CCl 4 ) intoxicated rats. Results: Among all extracts, ethyl acetate extract (EAE) possess potent antioxidant activity, viz., ferric reducing ability of plasma (abs = 0.379 ± 0.07), 2,2-diphenyl-1-picrylhydrazyl (inhibitory concentration 50% [IC 50 ]: 103.50 ± 2.05 µg/mL), OH• (89.33 ± 1.79 µg/mL), NO • (IC 50 129.34 ± 1.29 µg/mL), O 2ˉ (IC 50 62.03 ± 2.78 µg/mL), and inhibition of lipid peroxidation (110.05 ± 2.96 µg/mL). Treatment with EAE significantly increased the cell viability (IC 50 80.93 ± 1.02 µg/mL) by preventing CCl 4 induced cell damage in in-vitro HepG2 cell line. In case of both prophylactic and curative study, EAE extract significantly (P < 0.001) decreased CCl 4 -induced increased serum liver enzymes activities in CCl 4 -intoxicated rats, comparable to silymarin. Hepatoprotective potential further supported by pentobarbitone induced sleeping time and improved hepatic tissue histopathology. Study results suggest that antioxidant activity and hepatoprotective effect of EAE might be due to presence of polyphehols, viz., TFC (43.76 ± 0.78 Quercetin equivalent [QE] mg/g extract), TPC (194.16 ± 0.74 gallic acid equivalent [GAE] mg/g extract), and TTC (20.45 ± 2.31 GAE mg/g extract). Reversed-phase highperformance liquid chromatography analysis results showed highest quercetin content (32.8 ± 0.24 mg/g) in EAE. Conclusion: This study advocated that due to the presence of flavonoids, Z. xylopyrus leaves exhibited marked antioxidant and hepatoprotective activities.
