Pharmacologic mechanisms of serotonergic regulation of dopamine neurotransmission

Abstract: The neurotransmitter dopamine (DA) has a long association with normal functions such as motor control, cognition, and reward, as well as a number of syndromes including drug abuse, schizophrenia, and Parkinson's disease. Studies show that serotonin (5-HT) acts through several 5-HT receptors in the brain to modulate DA neurons in all three major dopaminergic pathways. There are at least 14 5-HT receptor subtypes, many of which have been shown to play some role in mediating 5-HT/DA interactions. Several subtypes… Show more

“…20 5-HT 2C receptors are also found at high density on cell bodies in the ventral tegmentum where they are believed to tonically inhibit dopamine release from dopaminergic neurons projecting to the frontal cortex. 21 Therefore, 5-HT 2C presents an intriguing but challenging target for therapeutics (i.e., agonism at 5-HT 2C in the mesolimbic dopamine pathway may reduce dopaminergic transmission and improve positive symptoms of schizophrenia).…”

“…It is likely that the predominant role of 5-HT 2A receptors in antipsychotic action is to modulate dopaminergic tone, particularly along the mesocortical pathway. 82,83 However, these studies also provide insight into why compounds with more complex pharmacologic profiles are likely superior to the 'magic bullet' approach in the treatment of complex diseases such as schizophrenia. 31,45 Serotonin 5-HT 1A receptors In addition to antagonism of the 5-HT 2A receptor, the agonist effects of clozapine on 5-HT 1A receptors have been postulated to contribute to its superior efficacy.…”

“…Indeed, aripiprazole, a D 2 receptor partial agonist, may owe some of its atypical properties to its net effect of weak D 2 receptor antagonism, 5-HT 2A receptor antagonism and 5-HT 1A receptor agonism. 32,38,85 As such, 5-HT 1A receptor modulation is most likely to play a role in regulating dopaminergic tone similarly to 5-HT 2A receptors, 83 thus contributing to atypicality. Particularly, 5-HT 1A receptor agonism has been suggested to enhance dopamine levels in the prefrontal cortex, 86 which may be related to the modest efficacy of many atypicals in treating the negative symptoms and cognitive dysfunction of schizophrenia.…”

“…For example, it is well documented that activation of the 5-HT 2A receptor can modulate dopamine levels or physiological responses mediated by dopaminergic systems (Alex and Pehek 2007;Lucas and Spampinato 2000;Pehek et al 2001;Vollenweider et al 1999;Yan 2000). In addition, it has been reported previously that pretreatment with 5-HT 2A agonists can potentiate the discriminative stimulus effects of amphetamine (Marona-Lewicka and Nichols 1997), methamphetamine (Munzar et al 1999(Munzar et al , 2002, and cocaine (Munzar et al 2002) in rats.…”
