Pharmacokinetics of Liposomal Amphotericin B During Extracorporeal Albumin Dialysis

Abstract: Extracorporeal blood purification techniques such as hemofiltration or albumin dialysis can exert a significant, but not easily predictable influence on plasma pharmacokinetics of antimicrobial agents. The effect of albumin dialysis on the pharmacokinetics of liposomal amphotericin B (AMB) and other lipid-formulated drugs has not been investigated so far. Therefore, plasma concentrations of liberated and liposomal AMB were measured in a patient, who obtained liposomal AMB for suspected invasive mycosis and req… Show more

“…Only two reports regarding the use of antifungals during AD were found in the literature, demonstrating that the influence of this technique on drug elimination appeared to be negligible [3,4]. We report the first pharmacokinetic study of anidulafungin during AD.…”

“…Thus, only small amounts of AMB, probably the liberated component, appear to be excreted via the bile during treatment with LAMB or ABCD. By contrast, in liver tissue, AMB concentrations of ~ 100 µg/g were measured, probably because of accumulation in reticuloendothelial cells [20]. demonstrated by microcalorimetry.…”

“…Pharmacokinetic data on antimicrobial agents-in particular on antimycotics-administered under AD are very sparse (8,9). AMB lipid preparations are clinically used for the treatment of invasive fungal infections in patients with a high risk of renal toxicity.…”
