Pharmacokinetics of Azlocillin in Persons with Normal and Impaired Renal Functions

Abstract: The pharmacokinetics of azilocilin, a new wide-spectrum antibiotic of the Nsubstituted group of ureidomethyl penicillins, was investigated in 10 subjects with normal and in 32 subjects with impaired renal functions. After intravenous injection of 2 g of azlocillin, serum concentrations of drug were measured microbiologically. The half-lives of azlocillin wete 47 ± 8.8 min in patients with normal renal function, and 293.3 min in patients with severely impaired renal function.

“…Serum levels remained well above the av erage reported MIC limits for P. aerugin osa strains [7,15] for at least 6 h in all pa tients. “…In patients with varying degrees of renal insufficiency, FIEGEL and BECKER (1978) found a linear relationship between inulin clearance rate and serum half-life on a log-log scale. In patients with end-stage renal disease, mean serum half-life is about 2 h on dialysis and twice that off dialysis.…”
