Pharmacokinetics of a novel extended half‐life glycoPEGylated factor IX, nonacog beta pegol (N9‐GP) in previously treated patients with haemophilia B: results from two phase 3 clinical trials

Abstract: N9-GP has the potential to shift previously treated haemophilia B patients from a severe/moderate disease state into a mild- or non-haemophilic range for most of the dosing interval, which is expected to reduce the number of bleeding episodes.

“…For all patients, FIX activity remained >0.05 IU/mL for up to 168 hours after single‐dose administration of N9‐GP 40 IU/kg . Geometric mean FIX activity 168 hours postdosing for single dose 40 IU/kg was 0.08 IU/mL (1‐6 years), 0.11 IU/mL (7‐12 years), 0.15 IU/mL (13‐17 years) and 0.17 IU/mL (≥18 years).…”

“…When steady‐state FIX activity profiles were predicted using a one‐compartment distribution model, patients treated with N9‐GP 40 IU/kg once‐weekly were predicted to have FIX activity >40% for 5.4 days per week in adolescents and adults (Figure ), and for 2.3 days per week in children …”
