Pharmacokinetics and pharmacokinetic-dynamic modelling of rocuronium in infants and children

Abstract: We have determined the pharmacokinetics and pharmacokinetic-pharmacodynamic relationship of rocuronium in infants and children. We studied infants (n = 5, 0.1-0.8 yr) and children (n = 5, 2.3-8 yr), ASA II, in the ICU while undergoing artificial ventilation under i.v. anaesthesia with an arterial cannula in situ and the EMG of the adductor pollicis muscle was monitored. Rocuronium 0.06 (infants) and 0.09 (children) mg kg-1 min-1 was given i.v. over +/- 5 min until 85% neuromuscular block was obtained. Arterial… Show more

“…Rocuronium, a nondepolarizing neuromuscular blocker with an action on presynaptic receptors, was used to produce paralysis of muscles below the cuff (5,45). The arm was positioned on a table and compressed by a rubber tube in a disto-proximal direction to minimize blood in the arm, and then the cuff was inflated.…”

“…~,s,6 Although mean recovery times, at equipotent doses, of rocuronium are similar, rocuronium shows prolonged recovery after repeated doses or an accumulative effect. 6 The clinical implication of this accumulative effect is uncertain. Neostigmine has been studied at doses between 30 and 80 ~ag.kg -I for reversal of rocuronium.…”
