Phagocytic activities in neonatal monocytes

Abstract: Monocytes play an essential role in cellular host defense as circulating phagocytes, as well as precursors of macrophages. We investigated the principal phagocytic activities in monocytes from cord blood of term infants by analysing adherence, random migration, chemotaxis, bactericidal activity, phagocytosis-associated chemiluminescence, production of superoxide anion (O-2) and generation of hydrogen peroxide (H2O2). All phagocytic functions of monocytes from neonates were shown quantitatively to be comparable… Show more

“…Chemotaxis of macrophages was evaluated using a modification of the Boyden chamber technique (Celloplex, Basel, Switzerland) as recently described (20). Pooled serum activated by zymosan (50 mg/ml Sigma, Munich, FRG) and FMLP I 0-5 M (Sigma) were used as chemoattractants.…”

“…The aliquots were added to ice cold HBSS (450 pl) and the cells immediately centrifuged at 200 x g for 2 min at 40" C and the supernatant was diluted to make agar pour plates for colony counting. Intracellular killing of S. aureus was measured as recently described (20).…”

“…Phagocytosis and intracellular killing by neonatal monocytes are comparable to those achieved by adult MNCs (25)(26)(27)(28)(29). However, these data are derived from stimulation studies using early-onset sepsis pathogens or latex particles; there are no analogous data on phagocytosis of SE by monocytes in preterm infants.…”
