We investigated relationships among insecticides and aquatic invertebrate communities in 22 streams of two soy production regions of the Argentine Pampas over three growing seasons. Chlorpyrifos, endosulfan, cypermethrin, and lambda-cyhalothrin were the insecticides most frequently detected in stream sediments. The Species at Risk (SPEAR) pesticide bioassessment index (SPEAR) was adapted and applied to evaluate relationships between sediment insecticide toxic units (TUs) and invertebrate communities associated with both benthic habitats and emergent vegetation habitats. SPEAR was the only response metric that was significantly correlated with total insecticide TU values for all three averaged data sets, consistently showing a trend of decreasing values with increasing TU values (r=0.35 to 0.42, p-value=0.001 to 0.03). Although pyrethroids were the insecticides that contributed the highest TU values, toxicity calculated based on all insecticides was better at predicting changes in invertebrate communities than toxicity of pyrethroids alone. Crustaceans, particularly the amphipod Hyalella spp., which are relatively sensitive to pesticides, played a large role in the performance of SPEAR, and the relative abundance of all crustaceans also showed a significant decreasing trend with increasing insecticide TUs for two of three data sets (r=0.30 to 0.57, p-value=0.003 to 0.04) examined. For all data sets, total insecticide TU was the most important variable in explaining variance in the SPEAR index. The present study was the first application of the SPEAR index in South America, and the first one to use it to evaluate effects of pesticides on invertebrate communities associated with aquatic vegetation. Although the SPEAR index was developed in Europe, it performed well in the Argentine Pampas with only minor modifications, and would likely improve in performance as more data are obtained on traits of South American taxa, such as pesticide sensitivity and generation time.

BackgroundPesticides are washed from agricultural fields into adjacent streams where even short-term exposure causes long-term ecological damage. Detecting pesticide pollution in streams thus requires the expensive monitoring of peak concentrations during run-off events. Alternatively, exposure and ecological effects can be assessed using the SPEARpesticides bioindicator that quantifies pesticide-related changes in the macroinvertebrate community composition. SPEARpesticides has been developed in Central Europe and validated in other parts of Europe, Australia and South America; here we investigated its performance in East African streams.ResultsWith minimal adaptations of the SPEARpesticdes index, we successfully characterized pesticide pollution in 13 streams located in Western Kenya. The East African SPEARpesticides index correlated well with the overall toxicity of 30 pesticides (maximum toxic unit = maximum environmental vs. median lethal concentration) measured in stream water (R² = 0.53). Similarly, the SPEARpesticides index correlated with the risk of surface run-off from agricultural fields (as identified based on ground slope in the catchment area and the width of protective riparian strips, R² = 0.45). Unlike other bioindicators designed to indicate general water pollution, SPEARpesticides was independent of organic pollution and highly specific to pesticides. In 23% of the streams, pesticides exceeded concentrations considered environmentally safe based on European first tiered risk assessment.ConclusionsIncreasing contamination was associated with considerable changes in the macroinvertebrate community composition. We conclude that pesticides need to be better regulated also in developing countries. SPEARpesticides provides a straightforward and cost-efficient tool for the required monitoring of pesticide exposure in small to medium streams.

The present work has focused on the application of the inverse-QSAR/QSPR problem for generating new structures of pesticides; this is in view of its extremely important and widespread use in several areas, particularly the agricultural field. For this reason, we implemented a methodology containing nine detailed successive steps that include a quantitative structure-activity/property relationship (QSAR/QSPR) study performed to develop a model that relates the structures of 190 pesticides compounds to their n-octanol-water partition coefficients (logk ow). We used the unique atomic signatures which represent the structures and acts as independent variables while the property (logk ow) as the dependent variable. The model was constructed using 130 molecules as training set, and predictive ability tested using 60 compounds. Modeling of logk ow of these compounds as a function of the signatures descriptors was established by multiple linear regression (MLR) using (LOO) cross-validation. As a result, a QSAR/QSPR equation with 14 atomic signatures was hereby obtained with a R 2 = 0.659273, Q 2 = 0.65617 and RMSE training = 0.930192, s = 1.37297 for the training set and in leave-one-out (LOO) cross-validation experiment set value, q 2 = 0.605676, RMSE LOO = 1.0936 respectively. In addition to all of the above, new structures have been generated for a range of pesticides that can be included as future search topics.
