Pest and disease resistance enhanced by heterologous suppression of a Nicotiana plumbaginifolia cytochrome P450 gene

Abstract: The functional role of the Nicotiana plumbaginifolia cytochrome P450 gene CYP72A2 was investigated in transgenic plants. N. tabacum plants transformed with a sense or antisense CYP72A2 construct exhibited diminished heights, branched stems, smaller leaves and deformed flowers. Western blot analysis revealed reduced levels of a 58 kDa protein corresponding to CYP72A2, suggesting that the CYP72A2 homolog was suppressed in the sense and antisense plants. Transgenic plants had increased resistance to Manduca sexta… Show more

“…In senescing leaves, leaf cytokinin levels are reduced, and external applications of cytokinins are well-known to delay tissue senescence. Transgenic plants that express the bacterial ipt (isopentenyl transferase) gene that catalyzes the synthesis of cytokinin exhibit delayed senescence (10,27,28) and increased insect and microbial resistance (29,30,32). In this paper, transgenic tobacco plants with constitutive or inducible promoters that overproduce cytokinins are utilized to analyze their response to infections with compatible and incompatible Pseudomonas spp.…”

“…Heterologous gene expression provides a means of examining the defensive role of metabolites in planta (Duan et al, 1996;Smigocki and Wilson, 2004;Zavala et al, 2004b). Introducing a Ser-type PI gene from potato (Solanum tuberosum) into rice (Oryza sativa) resulted in a new insect-resistant rice cultivar (Duan et al, 1996).…”

“…These variability's and the complex classification of CYTOCHROME P450 genes leads to diverse functions in plants [50]. Usually, these genes are involved in secondary metabolite biosynthesis [50] and defense responses against various abiotic and biotic stresses [50,51]. Plant CYTOCHROME genes activate during stress conditions and accumulate antioxidant flavonoids [12] to alleviate the negative effects of environmental stress [52].…”
