Perturbation of the mitochondrial import machinery by aggregation prone Tau affects organelle morphology and reduces neuronal complexity

Abstract: Protein import into mitochondria is an intricate and highly conserved process essential for organellar biogenesis, and maintenance of its structure and function. Defects in the import apparatus impact the assembly of the respiratory chain and ATP synthase complexes required for oxidative phosphorylation, compromising the ready supply of ATP to the cell. The consequences of reduced bioenergetic function are particularly severe for cells with high energetic demands such as neurons. However, relatively little is … Show more

“…Variants of the Huntingtin protein, the causative agent of disease, and the β-amyloid precursor APP have both been shown to associate with the TOM40/TIM23 complexes [12, 14, 52]. Indeed, in a parallel study we show that the a Tau variant linked with neurodegeneration associates with TOM40 and brings about TNT formation [53]. Therefore, these proteins may also act through binding to and impairing the import machinery, inducing TNT formation for aggregate efflux [5456].…”
