Cultivar Barangan is one of the famous banana plants that has good taste and price. The disadvantages of this banana is susceptible to Fusarium disease. Tissue culture techniques is one ways of plants propagation for this plant. This study is aimed to determine the effective concentration of benzyl amino purines (BAP) and indole-3 acetic acid (IAA) for optimal multiple bud clumps (MBC) induction in explants of Barangan Merah banana. The Research was carried out at the Laboratory of Tissue Culture, Faculty of Agriculture, Universitas Syiah Kuala. The research design was a completely randomized design, 3x2 factorial. BAP concentration was the first factor, i.e. 5, 10 and 15 mg L−1. The second factor was the concentration of IAA, i.e. 0.1 and 0.5 mg L−1. The results of this research showed that 96,29% explants of all treatments survived. Explant contamination was 11,11% occurred by bacteria and brownish exudate that spread to the medium. MBC are significantly increased at 16 days after induction at concentration of BAP 5 mg L−1 + IAA 0.5 mg L−1. All combinations of BAP and IAA concentrations have produced MBC explants (100%) at eighth weeks after induction. The concentration of 5 mg L−1 BAP + 0.5 mg L−1 IAA resulted the highest shoots number. Analysis of variance and Least Significant Different further test showed that the longest of shoot length was founding the concentration of 5 mg L−1 BAP compared to other treatments. Increasing concentrations of BAP 10 mg L−1and 15 mg L−1 resulted in significantly lower shoot length. It can be concluded that taken together the concentration of 5 mg L−1 BAP + 0.5 mg L−1 IAA is the best to effectively multiple bud clumps induction in explants of Barangan Merah banana through tissue culture technique.

A sufficient supply of healthy plants must be provided to balance the rising demand for bananas. A technique that can be used for propagation is tissue culture. Environmental factors and a variety of types are factors that affect explant growth. This research aimed to determine the effect of darkening duration on the growth of two banana (Musa sp.) varieties in vitro. In this study, a Factorial Randomized Block Design was implemented, considering two elements, namely the variety of bananas and the duration of darkening. The research showed that the time of shoot emergence started at 1.83 WAP in Cavendish grand naine treatment without darkening. The type of variety treatment had a significant effect on the number of leaves, and the best variety was Cavendish grand naine. The darkening duration treatment with four weeks of darkening increased shoot height significantly. An interaction between these two factors was observed in the number of shoots, shoot height, and number of leaves.

The existence of Sansevieria sp. as an ornamental plant that has many benefits preserved. However, conventional propagation requires a lot of materials and time long. Leaf cuttings are only able to produce 1-2 plants in 2 months, and stop when they are aged 5 months. While the separation of tillers (Tillering) only produces 2-3 plants from 1 clump for 5 months. Tissue culture can be the solution, but sterilization must be done to minimize contamination without killing explants. The research aims to find out optimal concentration of HgCl2 and duration of shaking on the sterilization of Sansevieria leaf explants sp.. The research design used a completely randomized design with 2 factors. first factor namely the concentration of HgCl2 with 3 levels (4%, 7%, and 10%,) and the second factor is duration shuffled with 4 levels (3, 5, 7, and 9 minutes). Each treatment includes a negative control performed at the time of explant sterilization in Laminar Air Flow. Data analyzed by test ANOVA or Kruskal-Wallis. Followed by the 5% BNJ test if it is significantly different. Results show that the concentration of HgCl2 has a significant effect on the parameters of the initial time of contamination, the mass sterility of explants, and percentage of browning explants, but had no significant effect on percentage of contaminated explants. Concentration that is too high can slow down the appearance of contamination, prolonging the sterile period, but causing browning of explants. Known that the duration of explant shaking in HgCl2 had no significant effect on all parameter. The use of 10% HgCl2 with a shaking duration of 7 minutes (H3P3) is known to be the most effective optimal for sterilization of explants Sansevieria sp. because it is able to reduce external contamination, prolongs the sterile period up to 12.33 HST, and does not cause too many explants browning that is 33.33%.
