Personalized immunoglobulin aptamers for detection of multiple myeloma minimal residual disease in serum

Abstract: Multiple myeloma (MM) is a neoplasm of plasma cells that secrete patient specific monoclonal immunoglobulins. A recognized problem in MM treatment is the early recognition of minimal residual disease (MRD), the major cause of relapse. Current MRD detection methods (multiparameter flow cytometry and next generation sequencing) are based on the analysis of bone marrow plasma cells. Both methods cannot detect extramedullary disease and are unsuitable for serial measurements. We describe the methodology to generat… Show more

“…In recent years, the emphasis on proactive healthcare diagnostics has steadily increased and shifted from reactive medical practices to preventative strategies [92][93][94][95][96][97]. Early disease prevention, a cornerstone of this movement, seeks to detect and address potential health issues even before they manifest as noticeable symptoms, where the current strides in the development of wearable devices equipped with TENGS hold great potential to revolutionize early disease prevention strategies by facilitating continuous, real-time health monitoring.…”

“…Aptamers are single-stranded oligonucleotides with a three-dimensional structure giving high affinity towards a specific target ( Figure 2 b). They are usually isolated by enrichment of single-stranded oligonucleotide wide libraries and reproduced through conventional DNA-synthesis techniques [ 112 ]. Like nanobodies, aptamers are characterized by low immunogenicity, high penetration capacity/high selectivity towards targets, and major solubility.…”
