Persistence of plasmid RP4 in Pseudomonas putida and loss of its expression of antibiotic resistance in a groundwater microcosm

“…We subsequently showed that the plasmid pB10 and the Pseudomonas putida host adapted during 1000 generations of growth in serial batch cultures to decrease plasmid cost and instability, thereby expanding or shifting the plasmid's range of favorable hosts. The instability of IncP-1 plasmids in a strain of Pseudomonas putida is in contrast to previous studies, where IncP-1 plasmids were stably maintained in Pseudomonas putida strains KT2440 and ATCC 12633 T for at least 50 or 68 generations in serial batch cultures (Schmidhauser & Helinski, 1985;Siddique & Figurski, 2002), in Pseudomonas putida mt-2(pWW0) for 56 days in contaminated groundwater (Jain et al, 1987), in Pseudomonas putida KT2442 for up to 26 days in aquifer (Bengtsson et al, 2004), and in Pseudomonas putida UWC1 for 20 days in soil (Götz & Smalla, 1997). A parallel study in our laboratory showed that plasmid pB10 was stable in strain Pseudomonas putida UWC1 for at least 210 generations (L. De Gelder et al, unpublished data).…”
