Perivascular Adipose Tissue Oxidative Stress in Obesity

Abstract: Perivascular adipose tissue (PVAT) adheres to most systemic blood vessels in the body. Healthy PVAT exerts anticontractile effects on blood vessels and further protects against cardiovascular and metabolic diseases. Healthy PVAT regulates vascular homeostasis via secreting an array of adipokine, hormones, and growth factors. Normally, homeostatic reactive oxygen species (ROS) in PVAT act as secondary messengers in various signalling pathways and contribute to vascular tone regulation. Excessive ROS are elimina… Show more

“…Oxidative stress is a cellular response caused by an imbalance between reactive oxygen species (ROS) production and antioxidant defenses, which can lead to cellular damage and in PLOS ONE some cases cell death [26,27]. Evidence is mounting that human OB is a state of chronic oxidative stress of the whole body since it perturbs the antioxidant defense system and reduces the activity of protective antioxidant enzymes in tissues [28][29][30]. Moreover, the chondrocytes and synovial fibroblasts of OA can also produce large amounts of ROS in response to biomechanical or biochemical stimuli, thus aggravating oxidative stress and cell damage in OA [31][32][33].…”

“…Recent attention has focused on the physiological impact of the PVAT on vascular physiology [11,[26][27][28][29]. The paradigm of paracrine signaling from the PVAT to the underlying vessel wall is particularly important in the regulation of vasoreactivity and vascular disease.…”

“…Obesity is characterized by oxidative stress and persistent inflammation [19], which can cause changes in the conformation and native properties of redox-sensitive enzymes of H 2 S metabolism and the development of deficiency of their cofactors and substrates. In particular, zinc, α-LA and thiosulfate are recognized antioxidants, and their need may increase under oxidative stress.…”

“…Ензим (кофактор) Схема реакції Ожиріння характеризується оксидативним стресом та персистуючим запаленням [19], що може викликати зміни конформації та нативних властивостей редокс-чутливих ензимів обміну H 2 S, розвиток дефіциту їх кофакторів та субстратів. Зокрема, цинк, α-ЛК та тіосульфат є визнаними антиоксидантами і за оксидативного стресу потреба в них може зростати.…”
