Periostin Supports Hematopoietic Stem/Progenitor Cells and Niche-Dependent Myeloblastoma Cells In Vitro

Abstract: Periostin (POSTN), the fasciclin family extracellular matrix protein also known as osteoblast-specific factor 2 (OSF-2), was previously reported to be required for optimal B lymphopoiesis in vitro. Now, our study first demonstrates the proof that POSTN might be a bona fide niche factor for both normal and malignant myelopoiesis, indicating that it is a niche molecule for hematopoietic stem cells and diverse hematopoietic precursor cells. The Mediator, composed of about 31 subunits, is a master transcriptional … Show more

“…Although a role for the periostin/ integrin avb3 axis for the support of hematopoiesis was elucidated, 23,44 periostin-deficient mice had an increased number of HSCs, characterized by increased cycling and faster recovery after sublethal irradiation. 23 HSCs in periostin-deficient mice exhibited poor repopulating ability on transplantation, similar to our data, but in vitro treatment of HSC with periostin decreased pAKT 1 HSC.…”
