Perinerin, a Novel Antimicrobial Peptide Purified from the Clamworm Perinereis aibuhitensis Grube and Its Partial Characterization

Abstract: A novel antimicrobial peptide was isolated and partially characterized from the homogenate of an Asian marine clamworm, Perinereis aibuhitensis Grube. This novel peptide, named Perinerin, was purified to homogeneity by heparin-affinity column and reverse-phase HPLC, and biologically tested with a MTS-PMS colorimetric assay. Perinerin consists of 51 amino acid residues and structurally appears to be highly basic and hydrophobic. It shows marked activity in vitro against both Gram-negative and Gram-positive bact… Show more

“…It has important applications in aquaculture [9] [10], fishing [11], and medical care [12], so the demand for P. aibuhitensis is high worldwide. However, the natural resources of P. aibuhitensis are depleting because of excessive harvesting and environmental degradation, so it has become F. Lv friendly.…”

AbstractIn this study, the effects of different salinity levels (9,12,15,18,21,24,27,30, and 33) on the growth performance, body composition, antioxidant indexes of Perinereis aibuhitensis (initial average mass, 20.4 ± 0.3 mg) and total nitrogen in the substrate were investigated. The survival rate, specific growth rate, feed coefficient, and protein efficiency ratio under different salinity levels were measured.

“…A 51-amino acid highly basic and hydrophobic peptide perinerin from the marine clamworm Perinereis aibuhitensis is an organism that is extensively used as bait in fisheries and aquaculture. Perinerin, a peptide that is constitutively present in the marine worm and whose sequence appears to be novel among all know antimicrobial peptides, was active against Gram-negative and Gram-positive bacteria as well as fungi (Pan et al, 2004). Antimicrobials peptides were screening both genomic and mRNA transcripts from a number of different species of flatfish.…”

“…Arenicins completely kill E. coli within 5 minutes at a concentration of 5 μM probably by membrane permeabilization [98] (Table 2). A second annelid peptide, named perinerin, has been purified from the clamworm, Perinereis aibuhitensis [9]. Perinerin contains four cysteine residues forming two disulphide bridges with the most abundant amino acid being arginine [9].…”
