Periadventitial Inducible Nitric Oxide Synthase Expression and Intimal Thickening

Abstract: Abstract-Positioning a silicone collar around the rabbit carotid artery induces a smooth muscle cell-rich intimal thickening. We investigated the localization of inducible nitric oxide synthase (iNOS) during thickening of the intima, the effect of iNOS inhibition on intimal thickness, and the effect of oxidized LDL (ox-LDL) on iNOS expression in the vessel wall. Collars were positioned for 18 hours or for 3, 7, or 14 days. Arterial cross sections were immunostained for iNOS, including naïve, sham-operated, and… Show more

“…Formalin-fixed carotid artery rings were paraffin-embedded and stained with hematoxylin and eosin or Verhoef's elastin. Immunohistochemical detection of macrophages (MO/RAM11; Dako Denmark A/S, Glostrup, Denmark) and SMCs (anti-␣-SMC actin, clone 1A4; Sigma-Aldrich) was carried out using an indirect peroxidase antibody conjugate technique (Kockx et al, 1992;De Meyer et al, 2000). The images were analyzed using a color image analysis system (Image-Pro Plus 4.1; Media Cybernetics, Inc., Silver Spring, MD).…”

Immunohistochemical detection was carried out using an indirect antibody conjugate technique (Kockx et al, 1992;De Meyer et al, 2000). The following primary antibodies were used: anti-␣-SMC actin (clone 1A4; Sigma-Aldrich) and MO/RAM11 (antirabbit macrophages; Dako Denmark A/S) on paraffin sections; anti-CD31 (JC/70A; Dako Denmark A/S) and anti-FITC (polyclonal antifluorescein isothiocyanate horse radish peroxidase conjugated; Dako Denmark A/S) on frozen sections.…”

“…Formalin-fixed carotid artery rings were paraffin-embedded and stained with hematoxylin/eosin (H/E) or Verhoeff's elastin. Immunohistochemical detection was carried out using an indirect antibody conjugate technique (Kockx et al, 1992;De Meyer et al, 2000). The following mouse primary antibodies were used: anti-␣-SMC actin (clone 1A4; Sigma-Aldrich) and anti-rabbit macrophage (clone RAM11; DakoCytomation Denmark A/S) on paraffin sections and anti-CD31 (JC/70A; DakoCytomation Denmark A/S) on frozen sections.…”
