Performance of the eazyplex® BloodScreen GN as a simple and rapid molecular test for identification of Gram-negative bacteria from positive blood cultures

Abstract: The LAMP-based eazyplex® BloodScreen GN was evaluated for the detection of frequent Gram-negatives directly from positive blood culture (BC) bottles. A total of 449 BCs were analyzed. Sensitivities and specificities were 100% and 100% for Escherichia coli, 95.7% and 100% for Klebsiella pneumoniae, 100% and 100% for blaCTX-M, 100% and 100% for Klebsiella oxytoca, 100% and 99% for Proteus mirabilis, and 100% and 99.8% for Pseudomonas aeruginosa, respectively. The time to result ranged from 8 to 16 min, plus abou… Show more

“…In turn, Bach et al [ 12 ] assessed the usefulness of the LAMP method-based assay in the identification of selected bacterial species and genes encoding ESBL enzymes from the CTX-M1 and CTX-M9 groups from positive blood cultures. The study included 449 positive blood cultures.…”

“…There have been many promising attempts to increase the speed of pathogen detection in positive BCs using culture-independent methods. Several PCR-based tests (e.g., BioFire ® FilmArray Blood Culture Identification from Biomerieux in Salt Lake City, UT, USA and Luminex ® VERIGENE ® from DIASORIN in Saluggia, Italy), matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF), or Loop-Mediated Isothermal Amplification (LAMP) based tests performed on a positive blood culture have shown to reduce pathogen detection time and optimize the antibiotic regimen [6][7][8][9]. The disadvantages of these methods are the requirement of a positive blood culture as well as the know-how and availability of the microbiology lab [5].…”

“…One promising technique is the loop‐mediated isothermal amplification PCR (LAMP) Eazyplex® (AmplexDiagnostics GmbH), which allows obtaining a fast result, in ~30 min, including the handling of the sample. Its usefulness has already been proved in the detection of other pathogens or targets, such as bacterial resistance genes or bacterial identification from blood cultures 17,18 . One of the advantages is that it can be used for one sample at a time, and it is easy to perform with few hands‐on‐time, being suitable for 24/7 on‐demand testing.…”
