“…Our results agree with recent investigations that reported increased stability of hPI in EDTA plasma and recommended this matrix for measurement of circulating concentrations [3,21]. Moreover some investigators have suggested that in addition to intact hPI, quantification of B-C junctional cleaved metabolites may have diagnostic value [11,12,27,39].…”

“…Accordingly, much effort has gone into developing improved immunoassay methods for quantitative Abbreviations: hPI, human proinsulin; RIA, radioimmunoassay; ELISA, enzymelinked immunosorbant assay; HPLC, high performance liquid chromatography; SEC, size-exclusion chromatography; cpm, counts per min. ଝ This manuscript reviews and summarizes work that was conducted at the determination of circulating concentrations of total, intact peptide [3,[18][19][20] and cleaved metabolites [21,22]. Early RIA methodology tended to detect both intact peptide and cleaved metabolites [23][24][25], while more recent sandwich ELISAs and related immunoanalytical methodologies have permitted the specific detection of intact peptide [3,19,20,21] and cleaved metabolites [22,23,27].…”

“…Резкое прекращение подачи инсулина или нарушение его абсорбции немедленно приводят к абсолютному дефициту инсулина в организме и угрожающим жизни метаболическим расстройствам. Всего через два часа после прекращения поступления инсулина из помпы в крови резко повышается содержание глюкозы и 3-гидроксибутирата [22,23]. Риск развития кетоацидоза при ППИИ, обусловленный техническими причинами, можно исключить или значительно снизить.…”
