Performance of a New Microfluidic Dengue NS1 Immuno-magnetic Agglutination Assay for the Rapid Diagnosis of Dengue Infection in Adults

Abstract: Dengue (DENV) infections are a public health concern worldwide and thus early diagnosis is important to ensure appropriate clinical management. The rapid diagnostic test (RDT) targets nonstructural protein 1 (NS1) detection and is the main tool used for diagnostic purpose. In this study, we evaluated the performance of a new rapid and semi-quantitative microfluidic DENV NS1 immuno-magnetic agglutination assay or IMA (ViroTrack Dengue Acute, BluSense Diagnostics, Copenhagen, Denmark). We studied 233 subjects co… Show more

“…1b). In contrast, the SD BIOLINE Dengue NS1 antigen kit was reported to show sensitivities less than 80% when compared with a newly developed serotyping NS1 ELISA system using DENV-2 and DENV-4 patient specimens in Thailand [37,38]. Previous evaluations of both the commercial and newly developed RDT NS1 detection systems showed false-negative results and various levels of sensitivity [17,24,[39][40][41].…”

“…Data from significant recent evaluation studies [15][16][17][18][19][20][21][22][23], using RT-PCR as reference, are summarized in Table 3. These data confirm the variable performance observed in earlier studies.…”

“…However, the proposed method emerges as an alternative powerful assay for rapid dengue diagnostic testing based on NS1 detection because it provides results within 15 min through a semiquantitative measurement and uses a small volume of sample (30 μL). A comparative study was performed by Wongsawat and colleagues (84), who achieved similar performance of the IMA assay compared to ELISA and better performance compared to immunochromatographic test for early and rapid diagnosis of DENV infections in endemic countries.…”
