Perforant path damage results in progressive neuronal death and somal atrophy in layer II of entorhinal cortex and functional impairment with increasing postdamage age

Abstract: In vivo model systems that can evaluate neuronal death, survival, and regeneration are critical to revealing basic mechanisms of neuronal response and developing strategies for CNS repair. We propose a distinct experimental model of CNS degeneration following lesions to the perforant path connecting the hippocampus and the entorhinal cortex. Within 2 weeks of a unilateral aspirative perforant path lesion, 30% of the ipsilateral entorhinal cortex layer II (ECL II) projection neurons had died with no change in t… Show more

“…Microscopy was performed by using a confocal microscope (Bio-Rad MRC1024UV). Quantification of ␤-gal-positive cells at each time point [2 days (n ϭ 3), 7 days (n ϭ 3), 14 days (n ϭ 4), 28 days (n ϭ 5), and 6 months (n ϭ 2)] was performed on every sixth section by using the optical fractionator procedure (MicroBrightField, Colchester, VT) as previously described (29,30). Statistical analysis was performed by multiway ANOVA followed by a Bonferroni post hoc analysis of means differences between groups (GraphPad, San Diego).…”

“…The volume of the septum was determined by the method of Cavalieri as described (31,32). A point-counting grid printed on an acetate sheet was placed over the monitor, upon which the whole septum was displayed from a low-power objective (ϫ4).…”

“…The inclusion of a significant portion of the axonal arbor of these neurons within the cortical gray matter in the form of local collaterals protects these cells from white matter lesions designed to avoid direct perikaryal damage. A recent model targeting entorhinal neurons with perforant pathway transections has produced, to date, inconsistent results (Cummings et al, 1992;Peterson et al, 1994). The rat olfactory cortex is an especially attractive area for investigations of pyramidal neuronal death for two reasons.…”
