Peptidomic Analysis of Urine from Youths with Early Type 1 Diabetes Reveals Novel Bioactivity of Uromodulin Peptides In Vitro

Abstract: Chronic hyperglycemia is known to disrupt the proteolytic milieu, initiating compensatory and maladaptive pathways in the diabetic kidney. Such changes in intrarenal proteolysis are captured by the urinary peptidome. To elucidate the early kidney response to chronic hyperglycemia, we conducted a peptidomic investigation into urines from otherwise healthy youths with type 1 diabetes and their non-diabetic peers using unbiased and targeted mass spectrometry-based techniques. This cross-sectional study included t… Show more

“…Targeted proteomics is gaining more importance as a diagnostic tool as it provides sensitive and accurate quantification of marker panels including native peptides or digested proteins [33]. The performance of the mass spectrometer is independent of biological products like antibodies making the technique independent of antigen interference, production variations, storage problems, and commercial availability.…”

“…1 ) is compatible with standard reversed-phase LC-MS/MS instrumentation available in most proteomics laboratories 41 . It is much less time- and labor-intensive than prior approaches, while also allowing the identification of 2-to-10-fold more endogenous human peptides from typical 1-10 mL urine samples 31,32,42,43 . This methodological advance allowed us to undertake a comparative analysis of the wheat-derived urinary peptidomes of patients with CeD and healthy controls.…”

“…Human primary renal proximal tubular epithelial cells (PTECs) were purchased from Lonza, Basel, Switzerland. The cells were cultured in DMEM/F12 media supplemented with 10% fetal bovine serum, 10 ng/mL epidermal growth factor, 5 μg/mL transferrin, 5 μg/mL insulin, 0.05 μM hydrocortisone, 50 units/mL penicillin, and 50 μg/mL streptomycin and incubated at 37 °C with 5% CO 2 as previously described [ 102 , 104 ]…”
