Peptidoglycan synthesis by partly autolyzed cells of Bacillus subtilis W23

Abstract: Partly autolyzed, osmotically stabilized cells of Bacillus subtilis W23 synthesized peptidoglycan from the exogenously supplied nucleotide precursors UDP-N-acetylglucosamine and UDP-N-acetylmuramyl pentapeptide. Freshly harvested cells did not synthesize peptidoglycan. The peptidoglycan formed was entirely hydrolyzed by N-acetylmuramoylhydrolase, and its synthesis was inhibited by the antibiotics bacitracin, vancomycin, and tunicamycin. Peptidoglycan formation

“…The solvents and biochemicals used were of analytical grade and obtained from BDH Laboratories (Poole, UK). Tunicamycin was a gift from Dr G. Tamura and stored in alkaline solution as described [17]. Moraprenol-PP-GlcNAc was supplied by Dr V. N. Shibaev and Dr S. D. Maltsev as a methanolic solution containing ammonium acetate (30.7 ninol GlcNAc-I-phosphate ml-I).…”

“…ATP is synthesized inside the cell, whereas lipoteichoic acid seems to be concentrated in the outer layer of the cytoplasmic membrane (21 and references therein). Alanylation of lipoteichoic acid may therefore be a transmembrane process such as has previously been documented for the biosynthesis of teichoic acid (4) and peptidoglycan (13,14) in B. subtilis.…”

“…We previously studied (22) the synthesis of peptidoglycan by intact cells undergoing partial wall autolysis and reached similar conclusions, although it was still possible that the wall-synthesizing complexes had been displaced from the membrane in the in vitro systems. Whereas displacement of the complexes does not occur in vivo, rotation of transmembrane complexes remains a plausible explanation for translocation in vivo; energy for such rotation or reorientation might arise from the electrocheniical proton gradient (AILH+) as previously suggested (22).…”

“…In a preliminary report (23) we showed that peptidoglycan formation in B. subtilis W23 is inhibited by the uncouplers DNP and CCCP; this occurs with an accumulation of the nucleotide precursors involved in peptidoglycan synthesis. However, since these uncouplers also affect the transport of N-acetylglucosamine (GlcNAc) (23) and have been reported to initiate autolytic activity (26), the study was extended to the synthesis of teichoic acid in vivo. A glycerol auxotroph (34) was used to allow maximum incorporation of glycerol into teichoic acid.…”
