Pentoxifylline Prevents Driamycin-Induced Myocardial Fibrosis and Apoptosis in Rats

Abstract: SummaryAdriamycin (ADR) is a potent antineoplastic agent, but long-term treatment is limited by its cumulative, life-threatening cardiomyopathy. Recently, a few reports have shown that pentoxifylline (PTX) might produce cardioprotection in cardiac dysfunction. Here, we investigated the protective effects of PTX on ADR-induced cardiomyopathy in rats. Male rats were randomly assigned either to saline, ADR (adriamycin, 5 mg/kg/week), or A (adriamycin, 5 mg/kg/week) + PTX (pentoxifylline, 50 mg/kg/day) groups. Aft… Show more

“…Similar to the report of Zang,29) during the experiment, myocardium injury induced by DOX treatment were aggravated in mice, but there were no signs of over-toxicity or deaths that happened. M-mode image of the LV displays the ventricular walls, LV cavity, and cardiac function measurements (Figure 1A).…”

“…For instance, PX administration inhibits myocardial apoptosis after adriamycin-induced dilated cardiomyopathy, by blocking the caspase-3 dependent apoptotic pathway. 33 However, in our study, it did not influence these parameters.…”

“…Immunohistochemistry: At the end of reperfusion, the heart was sectioned and small pieces of the heart were immersion fixed for 24 hours in 10% formalin solution. 16) Next, all of the samples were processed using a standard histological procedure. 12) After blocking in 5% BSA for 30 minutes, sections were incubated with anti-Sirt1 antibody (1:200) and anti-PGC-1α antibody (1:300) at 4°C overnight.…”
