Pentoxifylline for vascular health: a brief review of the literature

Abstract: Pentoxifylline is a methylxanthine derivative that has been used for several decades in the symptomatic management of intermittent claudication. For reasons that remain fairly obscure, this drug benefits blood rheology in a number of complementary ways: decreasing blood and plasma viscosity, lowering plasma fibrinogen while promoting fibrinolysis, and improving blood filterability by enhancing erythrocyte distensibility and lessening neutrophil activation. Anti-inflammatory effects on neutrophils and macrophag… Show more

“…Pentoxifylline is a non-specific PDE inhibitor used clinically for the treatment of intermittent claudication 197 and acts to suppress inflammation by inhibiting synthesis of pro-inflammatory cytokines and reactive oxygen species 77 . In mdx mice, pentoxifylline was shown to counteract hyperactive voltage-independent Ca 2+ channels in myofibers 77 , increase tetanic tension in the diaphragm, and reduce creatine kinase 198 .…”

“…Theobromine is mainly found in cacao and recent evidence shows that it has great potential as an antitussive agent [9]. Theophylline, aminophylline, doxofylline and diprophylline are used in the therapy of bronchial asthma and chronic obstructive pulmonary diseases (COPD) [8,10,11,12,13]. In particular, doxofylline and diprophylline are two novofyllines synthesized to have fewer side effects than other methylxanthines [11,12,13].…”

Pentoxifylline is used in the treatment of peripheral vascular diseases and in the management of cerebrovascular insufficiency and diabetic nephropathy [8,10]. Etofylline, another synthetic derivative, is used in lowering the blood lipid level [14].…”
