Seed viability is one of the most important physiological quality benchmarks of seeds. The use of appropriate germination media can affect the results of seed viability testing. This study aims to determine the optimal germination media in tobacco seed viability testing. This research was conducted at the Seed Laboratory of the Indonesian Sweetener and Fiber Crops Research Institute, Malang in January - March 2019. The research method used a two-factorial randomized block design (RAK). The first factor is tobacco seed varieties consisting of Kasturi in 2007, Bojonegoro in 2012 and Kemloko in 2014. The second factor is the seed germination media consisting of straw paper, cotton, towel tissue, paperboard and newspaper. The results showed that the Kemloko variety of tobacco seeds germinated in the towel tissue had the best seed vigor and viability percentage. Seed germination media with paperboard, towel tissue and straw paper can be used to test the viability of tobacco seeds because they have a percentage of simultaneous growth, percentage of the growth speed, percentage of germination, percentage of the growth potential maximum and normal seedling dry weight which are equally well

Kecipir merupakan tanaman yang mempunyai banyak manfaat dan berpotensi untuk dilestarikan. Karakteristik benih kecipir yang impermeabel terhadap air dan gas merupakan faktor yang diduga menyebabkan kecipir sulit untuk berkecambah. Penelitian ini bertujuan untuk mempelajari efektivitas metode pematahan dormansi dan pengaruhnya terhadap struktur benih kecipir. Penelitian dilaksanakan di Laboratorium Ilmu dan Teknologi Benih IPB dan Laboratorium Silvikultur SEAMEO BIOTROP selama 6 bulan. Metode pematahan dormansi dengan perlakuan larutan asam kuat HNO3 5% selama 10 menit dengan nilai daya berkecambah 93.33% dan keserempakan tumbuh 86.67% merupakan nilai yang nyata lebih baik dibanding kontrol sehingga metode ini dapat meningkatkan persentase daya berkecambah dan keserempakan tumbuh benih kecipir. Perlakuan larutan asam kuat HNO3 10% selama 15 menit dengan nilai indeks vigor 56.67% merupakan perlakuan yang dapat meningkatkan persentase indeks vigor benih kecipir. Perlakuan suhu 50 0C selama 10 menit dengan nilai kecepatan tumbuh 14.79% etmal-1 merupakan perlakuan yang dapat meningkatkan kecepatan tumbuh dan menghasilkan kecambah dengan akar yang lebih panjang dibandingkan kontrol maupun perlakuan lain. Perlakuan amplas tidak dapat meningkatkan viabilitas maupun vigor benih, bahkan menimbulkan pertumbuhan cendawan yang menyebabkan kemunduran benih.

Indigenous vegetables are part of Indonesia's biodiversity. Indonesia is among the three mega biodiversity after Brazil and Madagascar. Until now, the attention to indigenous vegetables is still lacking. The challenge is how to raise the potential of indigenous vegetables to compete with main vegetables and how to develop them. Related to the increasing need for food, efforts are needed to increase the development of indigenous vegetables, especially winged bean. For its development, post-harvest handling is required, one of which is drying. The problem faced was how the effect of temperature and drying time on the chemical, physical and organoleptic characterustucs of dry winged bean. Factorial randomized block design was used in the study. Research objected was to know effect of temperature and drying time on quality of chemical (vitamin C, ash, moisture content, protein, carbohydrate, fat, crude fiber), physical (rendemen) and organoleptic (color, taste, aroma, crispiness, appearance) of dried winged beans. The first factor was the drying temperature consisting of : 50°C and 60°C. Second factor was drying time, namely : 20 hours, 24 hours and 28 hours. The results showed that the interaction between temperature and drying time affected on vitamin C and yield. The best characteristic of dry winged bean was 60°C for 20 hours. As a suggestion: it was necessary to look for packaging that can maintain quality and shelf life.
