Penetration of the blood-brain barrier by peripheral neuropeptides: new approaches to enhancing transport and endogenous expression

Abstract: The blood brain barrier (BBB) is a structural and functional barrier between the interstitial fluid of the brain and the blood; the barrier maintains the precisely controlled biochemical environment that is necessary for neural function. This constellation of endothelial cells, macrophages, pericytes and astrocytes forms the neurovascular unit which is the structural and functional unit of the blood brain barrier. Peptides enter and exit the CNS by transport systems expressed by the capillary endothelial cells… Show more

“…BBB destabilization has been demonstrated to increase the incidence of stroke and neurodegenerative diseases as vascular dementia. 16,17 Moreover, the breaking of the BBB has been implicated in the transfer of the malaria parasite into the brain. 18 Immunoglobulin G (IgG) extravasation is commonly used as an index of BBB disruption.…”

“…BMECs are highly specialized structures due to their extensive tight junctions (TJs) between them (6). They play an important role in BBB function by restricting and precisely regulating the exchange of molecules between the peripheral circulatory system and cnS (7). consequently, as a result of the presence of the BBB, numerous drugs with a molecular weight of >400 Da cannot reach the lesions in the brain (8), severely reducing the therapeutic effect of drugs used in the treatment of cnS diseases.…”

“…The increased abuse of prescription opioids, including morphine, oxycodone, fentanyl, and the nonprescription opiate heroin has resulted in a severe public health crisis across large swaths of America [133][134][135]. In 2017, over two-thirds of drug-overdose deaths resulted from opioid abuse [136], and opioid-overdose-attributed deaths have tripled since the turn of the new millennium [137].…”
