Peneciraistin C induces caspase‐independent autophagic cell death through mitochondrial‐derived reactive oxygen species production in lung cancer cells

Abstract: Peneciraistin C (Pe-C) is a novel spiroketal compound isolated from the saline soil derived fungus Penicillium raistrickii. Our previous study showed that Pe-C exerted a potent cytotoxic effect on many kinds of cancer cell lines, especially on human lung cancer A549 cells. Here, we report the anticancer mechanisms of Pe-C in a variety of lung cancer cells. The results showed that Pe-C induced caspase-independent autophagic cell death and elevated mitochondrial-derived reactive oxygen species levels. Interestin… Show more

“…Recent evidence from several mammalian cells implicates the activation of autophagic death without triggering apoptosis under oxidative stress conditions4344. Although the induction of apoptotic PCD was observed in GCs with chronic H 2 O 2 incubation26, the current data suggest that the apoptosis-independent mitophagic death functions predominantly in the early stages of oxidative stress-induced GC injury.…”

“…21 Moreover, various experimental data show the induction of apoptosisindependent autophagic cell death under oxidative stress conditions. 22,23 However, whether autophagy is correlated with oxidative injury remains to be investigated in GCs.…”

“…5,6 Additionally, oxidative stress-activated autophagy initiates PCD without apoptosis induction in multiple types of mammalian cells. 22,23 Therefore, the inhibition of autophagy, which is associated with enhanced GC viability, might be a novel mechanism underlying the protective effects of FSH against oxidative stress.…”

“…Since a predominant number of NSCLC cases fail to be detected at the surgically resectable early stage of disease, therefore chemotherapy and radiotherapy remain the mainstay of the treatment for inoperable NSCLC patients. However, while there have been numerous drugs approved for use, they have often suffered from the limited clinical applicability due to the development of resistance by tumor cells and non-specific toxicity toward normal cells [ 3 , 4 ]. One such a drug, paclitaxel (PTX), a taxane plant product derived from the bark of the pacific yew tree Taxus brevifolia , has demonstrated a clinically significant activity against a broad variety of tumor types and has become a first-line treatment for NSCLC [ 5 ].…”
